Target

  • Function

    Mediates the uptake of glutamate into synaptic vesicles at presynaptic nerve terminals of excitatory neural cells. May also mediate the transport of inorganic phosphate.
  • Tissue specificity

    Predominantly expressed in adult brain. Expressed in amygdala, caudate nucleus, cerebral cortex, frontal lobe, hippocampus, medulla, occipital lobe, putamen, spinal cord, substantia nigra, subthalamic nucleus, temporal lobe and thalamus.
  • Sequence similarities

    Belongs to the major facilitator superfamily. Sodium/anion cotransporter family. VGLUT subfamily.
  • Developmental stage

    Expressed in fetal brain.

      Immunocytochemistical detection of VGLUT2 using antibody ab79157 on rat primary hippocampal neuronal culture. Fixative: Paraformaldehyde. Permeabilization: Triton 0.3% in 0.1X PBS. Blocking step: 10% Serum as blocking agent for 20 mins @ 24°C. Primary Antibody: 1 µg/ml for 1 hour @ 24°C in Donkey serum in 0.1%PBS plus Triton 0.3%. Secondary Antibody: Alexa Fluor® 568 Conjugation (1/1000). The submitted image shows ab79157 staining rat primary hippocampal neurons.

    Answer

    Thank you for your inquiry. The above mentioned antibodies are all tested to be specific for either VGlut1 or VGlut2. The sequence homology is only 73% between the two VGlut proteins and the antibodies are all tested to be specific for either VGlut1 or VGlut2. I suggest to purchase two antibodies raised in the same host, one against VGlut1 and one against VGlut2 and then maybe mix them to simultaneously probe for both proteins. But I have to admit that using them separately would be much cleaner and also better laboratory practice. I am sorry I could not provide a product that suits your purposes.
