      The wells were coated with ab200699 at 5µg/ml at 50µl/well overnight at 4°C, followed by a 5% BSA blocking step for 2h RT. Human IgG1 (ab90283) was then added starting at 100 µg/ml and plasma/serum at 1:50 and gradually diluted 1:4, 50µl/well for 2h. Ab201485 was then added at 1:10,000 dilution, 50µl/well for 2h. A HRP-streptavidin (ab7403) was used at 1:10,000 dilution for 1h.

    Answer

    Testing for glycosylation is not part of our standard QC and therefore we do not know the glycosylation status of this human IgG1.

    We do not perform a deglycosylation step in our purification and so we expect IgG1 to be glycosylated but we cannot guarantee this as it has not been assessed.

    Answer

    Neither of these proteins are recombinant. They are isolated from myeloma plasma, identified as IgG1 and IgG4 myelomas.

    These products are tested for possible contaminating IgGs by ELISA. Each of these products is at least 95% pure and by ELISA are at least 95% true to subclass.

    Thanks for the information. We decide to order one of the IgG1 mAb with kappa light chains, as you suggested. There are several types. Basically we need the mAb as pure as possible, and the storage buffer as simple as possible. Do you have human IgG1 kappa monoclonal? If not, I will order Mouse IgG1, kappa monoclonal. How pure is [MG1-45] - Isotype control (ab18447)?

    Tom is currently away from the office. Thus, I will be helping you. My name is Sybille.

    As for ab18447, it is a protein G purified antibody that comes in PBS without any preservatives. As per datasheet, ab18447 is 'LEAF Purified'. This is a low endotoxin azide free formulation containing no preservative. The antibody is 0.2 µm filter sterilized and the endotoxin level is <0.01 ng/µg protein.

    We have the human IgG1 protein (ab90283) that was purified human myeloma plasma and comes in 20mM PBS, 150mM Sodium chloride, pH 7.4 with 0.05% sodium azide. The nature of the light chain is kappa.

    There would be no human raised monoclonal antibodies as antibodies are not raised in humans. Human antibodies are only purified from human blood/plasma.

    The product ab90286 has kappa light chain however ab98893 has lamda light chain.

    The proteins are dissolved in different storage buffer and the concentration is different.

    These product are from human sources.

    But we do know there are 3 separate constant regions of the heavy chain and 1 found on the light chain.

    The information I have received so far from the lab is that they have not had this protein sequenced. They prepared the protein from the plasma of a single donor. It is not from a cell line.

    I'm still checking with them regarding the UniProt database entry. But I think it is safe to assume that the sequence is identical or almost identical, except for the occasional point mutation.

    The length of the guarantee, which promises a replacement or credit or refund if the product does not work as specified on the datasheet, is not the same as the expiration date.     IgG1 protein (ab90283) you will receive 1 mg at 3.41mg/ml, ¨so it will be in 293ul



    IgG2 protein (ab90284) you will receive 1 mg at 2.2mg/ml, so it will be in 455ul



    IgG3 protein (ab118426) you will receive 1 mg at 1.16mg/ml, so it will bein 862ul



    IgG4 protein (ab90286) you will receive1mg at a lot dependent concentration, between 1.09-1.12mg/ml, which will be between 917ul and 893ul
