    • Function

      Has both glyceraldehyde-3-phosphate dehydrogenase and nitrosylase activities, thereby playing a role in glycolysis and nuclear functions, respectively. Participates in nuclear events including transcription, RNA transport, DNA replication and apoptosis. Nuclear functions are probably due to the nitrosylase activity that mediates cysteine S-nitrosylation of nuclear target proteins such as SIRT1, HDAC2 and PRKDC (By similarity). Glyceraldehyde-3-phosphate dehydrogenase is a key enzyme in glycolysis that catalyzes the first step of the pathway by converting D-glyceraldehyde 3-phosphate (G3P) into 3-phospho-D-glyceroyl phosphate.

    Answer

    The Glutaredoxin 1 protein (Active), ab63231, is the Glutaredoxin 1 protein from Saccharomyces cerevisiae (SwissProt reference P25373).
    The activity of the protein was ascertained by monitoring its ability to oxidize NADPH. The activity was calculated to be 3X10^-3 micromol/min. The reaction mix contained NADPH (0.4 mM), GSH (1.0 mM), glutathione reductase (17 ug) and HED (1.4 mM) in 0.1 M Tris HCl, pH 7.4.
    A mixed disulphide between HED and GSH is formed within 2 minutes, and the reaction was started by the addition of 17 ug Grx1. The reaction was followed by the decrease in A340 due to the oxidation of NADPH.

    Thank you for contacting us and your interest in our products.

    The GAPDH protein ab82633 and ab77109 are both the full length GAPDH expressed in E. coli, however only ab82633 has been tested for its enzymatic activity. The specific activity was found to be 35.7 U/mg. One unit of activity of GAPDH is defined as the amount of enzyme required to convert 1 µmole of NAD to NADH per minute under standard assay conditions.
