Additional notes
Pre-activation is required for enzymatic assays. Please dilute Human MMP-9 to 100 µg/mL in TCNB buffer (50 mM Tris, 10 mM CaCl2, 150 mM NaCl, 0.05% Brij-35 (w/v), pH 7.5), and then add p-aminophenylmercuric acetate (APMA) to a final concentration of 1 mM. Please keep the enzyme with APMA for 8-24 hour at 37°C. Please note that the optimal treatment time may need to be determined empirically.
*100mM APMA stock solution should be prepared in DMSO. Please avoid adding high concentration APMA solution (>20mM) directly into the reaction as it tends to precipitate. A pre-dilution step is highly recommended. -

ReconstitutionIt is recommended to reconstitute the lyophilized protein in sterile deionized water to a final concentration of 100 µg/mL. Solubilize for 30 to 60 minutes at room temperature with occasional gentle mixing. Carrier protein (0.1% HSA or BSA) is strongly recommended for further dilution and long term storage.

Function
May play an essential role in local proteolysis of the extracellular matrix and in leukocyte migration. Could play a role in bone osteoclastic resorption. Cleaves KiSS1 at a Gly-
-Leu bond. Cleaves type IV and type V collagen into large C-terminal three quarter fragments and shorter N-terminal one quarter fragments. Degrades fibronectin but not laminin or Pz-peptide. -
