    Hematoxylin solution ab220365 is a high quality, ready-to-use solution of a commonly used formulation called Modified Mayer's hematoxylin (Lille's modification). 

    This hematoxylin solution is also available in H&E staining kit ab245880.

     Hematoxylin is a nuclear stain that is used as a counterstain for immunohistochemistry (IHC). Together with eosin, it forms the Hematoxylin and Eosin stain (H&E stain), one of the most commonly used stains in histology.

Answer

It’s not so much how to dilute the hematoxylin, but how strong you’d like the staining.


I like a really light stain (especially when doing IHC with HRP / DAB), so 5 seconds (or so) is all I expose the section to before transferring to running water. The degree of staining over any given time period will vary according to several parameters (for example, batch-to-batch variation, temperature, specimen type, etc).


If I were the customer, I take some of the specimens that they were planning to test, and stain for 5, 10, 15, 30 and 60 seconds, to see what degree of staining pleases them, then use this for their actual tests.
