Question

I recently bought ab7479 (bovine serum albumin), I will be using it for blockage in western blotting for monoclonal and polyclonal antibodies for phosphorylated antigens. Since I was using BSA in a different (powder) form, I couldnt be sure of the dilution necessary for this product. There is no data on the protein amount in the datasheet. WB protocol that I follow recommends to use 5% weight to volume preparation for the powder form. What is the recommended amount for this product? Thank you very much.

Thank you for your enquiry. The product ab7479 is not bovine serum albumin but bovine serum therefore it contains more proteins that just albumin. This product is typically used to reduce non-specific background staining which may be caused by a bovine secondary antibody during immunohistochemistry staining procedures. It may however work in western blotting. We typically recommend a 10% dilution (10ml serum plus 90ml PBST) as blocking agent for immunohistochemistry so I would recommend trying 5% and 10% (in TBST rather than PBST) and a one hour incubation.
