    Prestained Protein Ladder ab116029 is a three-color protein standard with 13 pre-stained proteins covering a wide range molecular weights from 3.5 to 245 kDa. Proteins are covalently coupled with a blue chromophore except for two reference bands (one green and one red band at 25 kDa and 75 kDa respectively) when separated on SDS-PAGE (Tris-glycine buffer).

    This product is also sold by Expedeon, an Abcam company as RunBlue™ TriColor Prestained Protein Ladder: NXA6050. This product was previously called Prism Ultra Protein Ladder (3.5 - 245 kDa).

    This prestained protein ladder is designed for monitoring protein separation during SDS polyacrylamide gel electrophoresis, verification of Western transfer efficiency on membranes (PVDF, nylon, or nitrocellulose) and for approximating the size of proteins. The ladder is supplied in gel loading buffer and is ready to use. Do not heat, dilute, add reducing agent before loading.

    Gel 1: Tris-Glycine (4-15%) SDS buffer; Gel 2: Bis-Tris (4-12%) MOPS buffer; Gel 3: Bis-Tris (4-12%) MES buffer. Pre-stained molecular weight standards have a differing mobility and as a consequence varying apparent molecular weight when run in distinct SDS-PAGE buffer systems. The variance in pH of alternative buffers affects the charge of the labelled protein standard and its binding capacity for SDS. The apparent molecular weight of this marker has been determined by calibration against an unstained ladder in each electrophoresis condition.

This Prestained protein ladder is a ready to use three-color (red, green, blue) protein standard with 13 prestained proteins covering an extra broad molecular weight from 4 to 245 kDa. Each vial contains 500 µl volume, sufficient for 200-500 gels. Figure: 1 µl per lane in SDS-PAGE gel and PVDF membrane.

Je peux vous confirmer que nos marqueurs de poids moléculaires, ab116027, ab116028 ou ab116029 ne contiennent aucune protéine avecun His tag.

Je tiens aussi à vous signaler, que des fois, en utilisant trop d'anticorps primaire, des protéines sont marquées de manière non-spécifique. Dans un cas comme ça, il suffit des fois de diluer l'anticorps primaire un peu plus.

Hallo,

Danke für ihre Antwort, bevor ich ihre ganzen Fragen beantworte hätte ich noch eine Frage, wäre es mgl das sie mir einen Marker für diese großen Proteine nennen? Ich konnte leider keine finden. Wir benutzen zum Transfer außerdem das System von Invitrogen das 7min dauert, mit Ponceau konnten wir zeigen das auch große Proeteine auf der Membran ware. Der Lysepuffer (1% Nonident P40, 500mmol/l Tris-Base (pH 7.6), 150mmol/lNaC). Zum Transferpuffer kann ich nicht soviel sagen da es keine gibt. Wir benutzen ja den iBlot ohne Puffer.

Frau Hoffmann ist im Moment im Urlaub und erst am Montag wieder verfügbar.

Leider bieten wir keinen Marker an, der höher als 225 kDa läuft. Allerdings habe ich einen Marker bis 300kDa bei Fermentas gefunden:

Da Sie ein sehr schweres Protein detektieren möchten, möchte ich Ihnen empfehlen, einen Ureapuffer zu verwenden. Ich habe Ihnen ein sehr interessantes Paper über die Löslichkeit von verschieden Proteinen hinsichtlich des Kompartiments und des Gewichtes an diese Email gehängt.
