Question

Dear Tracy, thank you for your information. We are interested in your products, in case you can provide the European Certificate( Country of Origin from Europe). As primary antibodies we choose ab2777 Prion protein PrP antibody [F 89/160.1.5]. And, if I am right, in order to test 2500 samples, we need 5 packs of antibody? Is it true? But we need also to examine caprine samples, so we can`t use biotinylated anti-mouse IgG raised in Goat. As secondary antibodies we prefere rabbit or donkey polyclonal to mouse IgG (biotin). We found two donkey polyclonal (ab7060 and ab6819) and three rabbit (ab6727, ab7074 and ab5761). Can you propose the most suitable secondary antibodies (minimal staining background), conjugated with biotin, and how many packs are required for 2500 samples? We are not sure, whether ab9135 (Endogenous Enzyme blocker) corresponds to peroxidase blocking solution- hydrogen peroxide solution per methanol 1:100. If it is the same, then it suits us.. We are interested in ab3385 PBS concentrate, in HRP chromogen kits (ab8184 or ab5814), in ab8193 Clarion mounting medium as well.. Maybe ab9909 TissueBond slide adhesive would suit us, if it is APES (3-amino-propyl-triethoxysilane), but maybe you can propose already coated 2500 slides? How long can stay with ab9909 coated slides? And final, can you propose separatedly Streptavidin peroxidase conjugate and Mayer`s hematoxylin in required quantity? In summarizing, we are interested in all these products in the required quantity, necessary for investigation of 2500 samples . At least 6 months warranty should be supplied and must have Certificate Country of Origin from Europe. Thank you and look forward to hearing from you soon. Your's sincerely, Irena

Answer

Unfortunatley we do not have country of origin certificates for our products nor do we offer six months warranty. Our guarentee is that an antibody should work as stated on the data sheet or we will refund or replace it. I also cannot hazard a guess at how much antibody you need as every assay is individual. The recommended dilutions are useful as a guide but it is due to each researcher to produce the optimum dilutions for their assay. For ab2777 the optimun dilutions are 3-5ug/ml and the pack size is 200ug. So you should have 40mls of antibody solution but I do not know how much you intend to add to each of your samples. Again I am unable to recommend the smost suitable secondary because every assay is different and what suits one researcher may not suit another. I suggest you read the datasheets for these antibodies and check for reviews from previous customers as well as any information on cross reactivity and specificity. ab9135 is used to suppress endogenous phosphatase and peroxidase-like enzyme and should be used as 1ml of conc endogenous enzyme blocker to 5ml of alcohol. ab9909 is very similar to APES and can be used in the same way. Once coated the slides can be stored for future use. We have a number of slides available on our website but for custom slides we suggest you use a company called Biochain. For all of our products we always put all the information we have upon the datasheet, therefore I suggest you read these carefully. We cannot recommend products on an individual basis due to the variation in the tests reasearchers use. For that reason also we can give recommended dilutions but not predict how much a researcher would use. I am sorry that these answers are a little vague but hope they will be of assistance to you.

Question

Hi, was this antibody adsorbed against Armenian hamster or Syrian Hamster or both? I am look it use it as a secondary in Western Blotting after immunoprecip. with an Armenian Hamster monoclonal and blotting with a polyclonal mouse serum. However, my protein of interest runs very close to the Ig heavy chain at 50kDa. Thus I am picking up my IP antibody with my current secondary and am looking for one that won't cross-react between the two species.

Answer

This antibody is pre-adsorbed against Golden Syrian Hamster serum proteins. The cross reactivity to this species is very minimized. Cross-reactivity to Armenian hamster is unknown.
