I am looking for a HRP conjugated anti-mouse antibody to be used in ELISA. The most important thing is that it will have the least cross reactivity to rat immunoglobulins. I see that you have many such antibodies. Which one would you think is the most suitable?

Answer

Thank you for your enquiry. I am sorry to hear that you have been obtaining high background with this antibody. Following contact with the source of this antibody I have determined that the antibody is affinity purified, then cross adsorbed using solid phase adsorption. This means the affinity purified antibody is passed through column(s) bound with listed species of IgGs. The antibodies which cross react with the listed IgGs will bind to the column(s), thus being removed from the final product. The major background in IHC and FACS is from Fc receptors on the cell surface. I think the best way around this problem is to use antibodies specific to the F(ab')2 fragment instead of whole IgG. e.g. ab5766 Mouse IgG antibody H&L - F(ab)2 Fragment Texas Red conjugated, Goat, FACS, IM. I do also think that it is worth inhibiting the peroxidase activity of this enzyme to ensure that the signal that you are observing is not due to the HRP substrate reacting with the endogenous peroxidase activity. I have determined from the source of this antibody that a good starting dilution is 1:1000. I hope this information helps, please do not hesitate to contact me should you require further assistance.
