Question

Nice talked with you on the phone about the our order 1773Ab. We are so sorry about the results and send the results to you (see attach file). The protocol of Dot blot are following: 3 ul of serum and TK1puri. were dot on the NC membrane and then blocked in TBST with 10 % non-fat milk for 4 hours . The presence or absence of the primary (anti-thymidine kinase1,TK1) antibody (we produced) was incubated at +4 C over night After incubation with a biotinylated secondary antibody (from Abcom 7113 6000x dilution or from Jackson 10000x dilution) was added for one hour at room temperature. The membrane was then incubated in TBS buffer with avidin-HRP-streptavidin (from Amersham). Finaly ECl detection was reaction 1 min and then exposed to X-ray film. The intensity of a single spot on the film was determined by a Laser Densitometer. From the intensity of the rhTK1 . Results: Now we found the higher cross-reaction between Abcom 7113 and serum sample, did not find for the second Ab from Jackson. See fig. We really hepe that you will give an answer soon.. And We aslo found the 7113 Ab without 50% glycerol for stored at -20 C. We are sorry about te results.

Answer

Thank you for contacting us for technical support and for sending us all this protocol information and an image, it is very useful to understand your problem. The antibody ab7113 has not been tested for DotBlot so we do not know if it will work in this application and we cannot guarantee it will. I think the problem you are having may be due to the blocking: I would recommend trying 5% milk or 5%BSA in TBST to block, only for 1hour, then add the primary and then the secondary antibodies (diluted in TBST only, no milk). This will help remove the non specific binding. I hope this suggestion will help you, please let me know if I can be of further assistance,
