Poor detection of mouse IgG in rat brain with significant background

Poor
Abreviews

abreview image
Application
Immunohistochemistry (PFA perfusion fixed frozen sections)
Image is a subsection of rat brain with ventricle and white matter tracts visible. In sections with primary antibody, a 1:125 dilution of secondary antibody resulted in very weak signal. In this image, only secondary antibody was added at the same dilution (i.e. secondary-only control) and developed a strong background signal of white matter tracts. Cell nuclei were counterstained with DAPI (blue).

Asked on Mar 21 2012

Answer

Yes, you can use ab6947 with ab54427. The antibody that recognizes IgG will recognize all IgG subtypes, whereas the IgG1, will only recognize IgG. Therefore, if you are not intending to use any other mouse primary antibodies at the same time then the IgG antibody will work well, if however you would like to use other mouse antibodies, then I would recommend the IgG1.

Please let me know if there is anything else I can help you with, and if you can provide me the address, I can generate a quote.

Question

I selected the following antibodies from the ABcam catalog:

Primary:

Anti-CD63 antibody [AD1] (FITC)

ab108949

$337.00

Anti-CD62P antibody [Psel.KO.2.7]

ab54427

$337.00

Anti-CD42d antibody [1C2]

ab95780

$337.00

Secondary:

Goat anti-Mouse IgG H&L (Cy5.5 ®)

ab6947

$185.00

Goat anti-Mouse IgG1 heavy chain (FITC)

ab97239

$165.00

Goat polyclonal to Armenian Hamster (TR)

ab5743

$165.00

Isotype controls:

MOUSE IgG1 antibody [MOPC-21] (FITC)

ab106163

$325.00

Mouse IgG1 [ICIGG1] - Isotype Control

ab91353

$315.00

Armenian Hamster IgG monoclonal [HTK888] - Isotype control

ab18479

$325.00

Two questions:

For the secondary antibody, will the first one (ab6947) work against ab54427, or do we need to buy the second one (ab97239)? One is against IgG, the other one is against IgG1. ab54427 is IgG1.

If you could double-check the selection for compatibility (e.g., correct isotype controls) and then ask the sales department to offer us a quote, that would be great.
