    HRP or AP conjugated to a polymer backbone, which is collectively conjugated to the antibody. The end result is two antibody subsets: 1) Multiple HRP molecules linked to goat anti-rabbit antibodies. 2) Multiple AP molecules linked to goat anti-mouse antibodies.

    This product is specially designed for a rapid double stain procedure. The overall staining procedure can be done in 4 steps and in less than two hours. Avidin-biotin blocking procedures are not necessary when using this product. This new and innovative AP and HRP polymerization technology provides a significant increase in staining sensitivity when compared to other conventional AP or HRP conjugated secondary antibodies.

    This product is provided in a pre-diluted, ready-to-use format. Procedure (adapt in-house protocols to accommodate): 1) A primary antibody cocktail with a rabbit monoclonal/polyclonal and a mouse monoclonal/polyclonal antibody must be applied for the standard duration. Rinse in TBS. (DO NOT USE PBS). 2) Apply ab2892 for 20 - 30 minutes then rinse in TBS (DO NOT USE PBS). 3) Chromogen 1: Develop AP using suitable chromogen. Rinse in TBS. 4) Chromogen 2: Develop HRP using suitable chromogen. Rinse in TBS. Equally suited to both manual and automated immunostaining proceedures. This product has not been tested in any application requiring further dilution of the antibody and therefore it is not recommended for use in such applications.

Thank you for your enquiry. The TBS buffer than we recommend is a general TBS buffer (pH7.6). Please find below the ingredients for the buffer. TBS 10x (concentrated TBS) - 24.23 g Trizma HCl - 80.06 g NaCl - Mix in 800 ml ultra pure water. - pH to 7.6 with pure HCl. - Top up to 1 L. Answer

Thank you for your enquiry. Yes, this can be used to perform single staining. However, I would recommend that your customer uses an HRP or AP conjugated secondary antibody together with a suitable chromogen as this would be a more simplistic approach. For example: Goat anti-rabbit IgG antibody (ab7090). However, I appreciate that we do not sell an AP conjugated anti-mouse secondary and if this was the approach it may be necessary to use this kit for detection purposes.

I am interested in knowing the reason on why you suggested to use TBS instead of PBS in the application note for catalog# ab2892 (mouse + rabbit double staining secondary antibody). Do you typically get better results with TBS as compared to PBS? If we are using a phosphor antibody, would you recommend using TBS for both the primary and the secondary antibody stain?

Thank you for your enquiry and your interest in our products. We normally suggest TBS instead of PBS for IHC staining. For details on IHC and antigen retrieval, please click on the Resources section on the top panel of the Abcam homepage, you might find some useful information there. As a general rule, for phosphor antibody we would suggest blocking the non-specific binding sites with 1%BSA rather than with milk. We hope this will help.
