This antibody may cross react with IgG from other species.

    ICC/IF image of ab6046 stained HeLa cells. The cells were 100% methanol fixed (5 min) and then incubated in 1%BSA / 10% normal goat serum / 0.3M glycine in 0.1% PBS-Tween for 1h to permeabilise the cells and block non-specific protein-protein interactions. The cells were then incubated with the antibody (ab6046, 1µg/ml) overnight at +4°C. The secondary antibody (red) was ab150083 Alexa Fluor® 647 goat anti-rabbit IgG (H+L) used at 2µg/ml for 1h. DAPI was used to stain the cell nuclei (blue) at a concentration of 1.43µM.

IF was performed on mouse cell-line. Fixation with 4% PFA for 1hr at RT, followed by blocking with 3% FCS solution. Primary antibody used from other company, diluted 1:1000 in blocking solution. Incubated for 16hr, 4oC. Secondary antibody diluted 1:200 in blocking solution, incubated for 1hr at room temperature.

30 um sections from PFA-perfusion fixed adult animals were cut on a cryostat and blocked for 1 hour in 2% goat serum. Primary antibody incubation was carried out in 0.3% PBST overnight at 4 degrees.

I tried many different product to get secondary antibody for immunofluorescence staining better and more clear but failed then I bought abcam Goat Anti-Rabbit IgG H&L (Alexa Fluor® 647) preadsorbed (ab150083) in kidney cells, as recommended in their datasheets and I never had any problem with it. I think its great product to use. I think this is best secondary antibody to detect immunofluorescence during in vivo studies

Question

I am going to stain mouse spleen cells with CD4-Alexafluor (FL-1) and IL4-PE (FL-2). The host for these antibodies are rat or mouse. It would be good if I get anti-BDKRB2 antibody as APC (FL-4) and the antibody should not cross-react with rat antibody.

I have also included two of our Alexa 647 antibodies (https://www.abcam.com/goat-polyclonal-secondary-antibody-to-rabbit-igg-hl-alexa-fluor-647-pre-adsorbed-ab150083.html & https://www.abcam.com/donkey-polyclonal-secondary-antibody-to-rabbit-igg-hl-alexa-fluor-647-pre-adsorbed-ab150063.html) which have emmision/excitation in the same range as APC. I have included these because they have lower cost, greater size and have been pre absorbed against rat, mouse and human. All three products are guaranteed under our AbPromise for use in flow.
