    Sections were double labeled for different PAK isoforms, together with calponin (marker for smooth muscle cells), pan-cytokeratin (marker for epithelial cells of glands), or tyrosine hydroxylase (TH, marker for catecholaminergic nerves).     ab11175 staining Histone H2A.X in mouse testis tissue sections by Immunohistochemistry (IHC-P - paraformaldehyde-fixed, paraffin-embedded sections). Tissue was fixed with paraformaldehyde, permeabilized with 0.1% Triton X-100, and blocked with 4% BSA for 2 hours at 37°C. Antigen retrieval was by heat mediation in a citrate buffer. Samples were incubated with primary antibody (1/100 in PBS/4% BSA/0.05% Triton X-100) for 48 hours at 4°C. A Cy5®-conjugated goat anti-rabbit IgG H&L preadsorbed (ab6564) (1/300) was used as the secondary antibody.

Worked well as a secondary antibody to detect a rabbit primary antibody by flow cytometry.
Used at 1:100 dilution in FACS buffer. Stained T cells from canine with this antibody. The red peak identifies the population that is recognized by the secondary only while the right peak is the population identified by the primary + secondary.

Both of these antibodies will be guaranteed for your purposes, and we would expect them to give equivalent signal and specificity. The main difference between the two is the recommended dilutions for ICC/IF. You should be able to get more tests per vial by using ab6564.

We recommend using this antibody in FC 1"10 - 1:100 dilution. We tested permeabilized HeLa cells and the histograph can be found on the datasheet: https://www.abcam.com/Desmin-antibody-Y266-ab32362.html

This is the recommended protocol:

Flow Cytometry Protocol for Intracellular Staining

1. Solutions and Reagents

1.1. 1X PBS

1.2. Blocking buffer: 0.5% BSA in 1X PBS

1.3. 2% paraformaldehyde (1% solution - optional for storing samples)

1.4. 1X FACS permeabilizing solution (BD Biosciences cat. #340973)

1.5. Fluorescently-conjugated secondary antibody (various forms)

2. Protocol

2.1. Collect 1x106 cells/sample.

2.2. Wash cells once with blocking buffer.

2.3. Fix cells with 2% paraformaldehyde and incubate at room temperature for 10 min.

2.4. Wash cells once with blocking buffer.

2.5. Add 0.5 ml 1X FACS permeabilizing solution and incubate at room temperature for 10 min.

2.6. Wash cells once with blocking buffer.

2.7. Incubate cells in blocking buffer for 30 min at room temperature.

2.8. Add primary antibody at the appropriate dilution and incubate for 30 min at room temperature.

2.9. Wash twice with blocking buffer and incubate with fluorescently-conjugated secondary antibody for 30 min at room temperature.

2.10. Wash cells twice with blocking buffer.

2.11. Re-suspend cells in 1X PBS and analyze on flow cytometry. Samples can be kept in 1% paraformaldehyde at 4 °C overnight.

I guess I am not clear what the "testing discount application" actually means.
Do we purchase the antibody at full price and get 1 for testing and 1 for usage?
My concern is, if we test it and it is not effective in the desired application, then we dont want it.

I can confirm that ab6564 is not yet tested experimentally inWB and therefore we cannot guarantee that ab6564 works in WB. But it is very likely to work in WB.

As my colleague who is not in the office today explained: the testing discount program requires you to purchase the antibody that you would like to test (here in WB). After submission of the Abreview the code will be valid for any product from our catalog.

Even if the tested antibody is not suitable for WB you will receive a free product of your choice to the value of the tested antibody.

You will also receive a free product of your choice from our catalog if the tested antibody works.

Unfortunately, we do not have any anti rabbit secondary antibodies that are conjugated to Cy5 and are tested in WB in our catalog at this moment in time.

Thank you for your enquiry. Cy5 emits the color red (it is located at the Far-Red end of the spectrum).
