    Thank you for contacting Abcam regarding ab96885.


    The conjugate should be relatively stable during storage in the dark. In our hands, we've read the same stained tissue every min for 10 min, without noticeable loss in intensity. Similarly, we stored the covered tissue at RT for more than a month and found no significant difference in fluorescence intensity.


    For your specific situation, the fluorescence intensity should not be noticeably different after 72 hours. However, if you are performing tight quantitative measurements, it is always better to make preliminary tests, just to make sure.

    This customer has purchased ab96885 (Goat anti-Rabbit IgG H&L (DyLight® 594) secondary antibody), ab96871 (Goat anti-Mouse IgG H&L (DyLight® 488) secondary antibody) and has conducted the ICC several times. This customer indicated she will observe the PE fluorescence when she wants to observe the FITC fluorescence, but this state doesn’t happen when she wants to observe the PE fluorescence, therefore, she would like to ask for your help to find out any possible problem and also help her to modify her experiment step, could you please offer any suggestion to her?

    Having reviewed this case, I would like to offer some suggestions to help optimize the results from abab96885 Goat anti-Rabbit IgG H&L (DyLight® 594) secondary antibody. I would also appreciate if you can confirm some further details:

    1. What kind of filter or lamp are you using? Unfortunately, DyLight® 488and DyLight® 594 do overlap: a little bit in the DyLight® 488 spectrum, but more important in the DyLight® 594 wavelengths, which corresponce with your findings. Check with the manufacturer of your microscope if their filters are suitable for those dyes.

    2. In order to check if the ab96885 is damaged I would suggest to perform an ICC experimentwith only one primary- secondaryreaction. This way you canexclude the annoying "bleed-over".


    Should the suggestions not improve the results, please do let me know.

    In the event that a product is not functioning in the species and applications cited on the product datasheet (and the problem has been reported within 6 months of purchase), we would be pleased to provide a free of charge replacement, credit note, or refund.

    Question

    We have an antibody (ab96885) that was stored at the back of our fridge and
    the fridge malfunctioned and iced up at the back, causing everything to
    freeze. On the storage conditions for this antibody, it states that the
    antibody should be stored at 4 degrees. Do you think that freezing it will
    adversely affect it? Most antibodies we have are stores at -20 and they are
    fine.

    This secondary antibody is Dylight 594 conjugated, which is why we recommend storing it at 4C. Freezing this antibody can affect the fluorophore. You can try it with a rabbit primary antibody to see if it can be detected and the fluorophore is still viable.


    At this point it is difficult to say if it will work or not without testing it because we haven't stored it frozen in our labs. I am sorry that this information could not be more helpful, but please contact us with any other questions.

    As he mentioned, if western blotting and IHC are listed as applications on the datasheet, and rat and mouse are listed, then the antibody will be guaranteed for those applications and species and a replacement or credit or refund is available in the event it fails.     We do not sell Alexa-conjugated secondary antibodies. However, we have launched a new family of DyLight® fluorochrome- labelled secondaries which exhibit higher fluorescence intensity, photostability and water solubility and remain fluorescent from pH 4 to pH 9.

    Additionally, the water solubility of the DyLight® fluorochromes allows a high dye-to-antibody ratio to be achieved without causing precipitation of conjugates. You customer can find some further useful details on DyLight® secondary antibodies at our home site:
