    The paraffin sections were deparaffinized and rehydrated, followed by antigen retrieval using sodium citrate buffer (pH 6). The sections were then incubated with 3% hydrogen peroxide to block endogenous peroxidase activity for 10 minutes at room temperature. After 3 washes with TBS, the sections were incubated with 2% bovine serum albumin for 1 hour at room temperature.

    Slides were then incubated separately with rabbit anti-Ki67 and rabbit anti-cleaved caspase 3 overnight at 4°C. The slides were washed 3 times and incubated with goat anti-rabbit HRP polymer (1:1, Abcam ab214880) for 2 hours at room temperature. Samples were washed thrice and then developed, counterstained with hematoxylin and then dehydrated before mounting the slides with permount.

Immunohistochemistry (Formalin/PFA-fixed paraffin-embedded sections)
The FFPE mouse tumor xenograft tissue sections were deparaffinized and undergo antigen retrieval with Citrate pH6 buffer in pressure cooking for 10 minutes. The sections were incubated with Rabbit anti-Ki-67 primary antibody overnight. After washes in TBS and TBST, the sections were incubated with 2x dilutions of Goat anti-Rabbit IgG (HRP polymer) secondary antibody. 2% BSA in PBS were used as blocking reagent as well as antibody diluent. DAB detection kit from Vector lab was used for detection.
