Combined with anti-CD3 antibody (ab16669) and anti-CD19 antibodies (ab25232) and Goat anti-Rabbit IgG H&L (DyLight® 488) secondary antibody ab96895:

Following fixation in in 4% paramformaldehyde in PBS for 15 min, slides were blocked with 10% goat serum for 30 min and washed 3 times with ice-cold PBS, and incubated with mixture of anti-CD3 (ab16669) and anti-CD19 antibodies (ab25232), diluted 1:200 in PBST (1× PBS with 0.05% Tween-20), 1% BSA for 1 hour in a humidified chamber at room temperature.
Slides were washed 4 times in PBS (5 min each) and incubated for 1 hour in dark at room temperature with mixture of secondary antibodies: Goat DyLight® 488-conjugated anti-Rabbit (ab96895) and Goat Cy3®-conjugated antibody to Rat IgG - H&L (ab98416), diluted 1:200 in PBST, 1% BSA.
Slides were washed 3 times with PBS in dark (each wash for 5 min) and counterstained with 0.1 µg/ml DAPI for 5 min and finally rinsed with PBS, left 10 minutes for air-drying in dark and finally mounted.
CD3 antigen appears green CD19 red and the nuclei are stained blue.
Fluorescent microscope pictures at 10x.
