Phoned because the customer is seeing very high background on the blot everywhere but the lanes which contain sample. Still getting good, clean signal on sample but was curious about this response.

Answer

Thank you for contacting us.

I have had the opportunity to speak with my collegues regarding this issue.We have seen similar patterns when the gel is over-heating during transfer or contaminants are present in the transfer buffer or equipment. So we definitely recommend using all new buffers and doing your transfer a lower/colder conditions.

I also wanted to suggest using a no primary antibody control to see if you experience the same results.

Question

Recently, I bought 5 antibodies from abcam, which are: 1 x GABARAPL2 antibody(ab15858) 1 x CREB antibody (ab5803) 1 x Leptin Receptor antibody (ab7211) 1 x GABARAP antibody (ab1133) 1 x Chicken IgY (H&L) antibody (ab6753) The order reference number is 66504 (PON: L-15) I receive them yesterday and 3 of them were OK, but I think that there was some kind of trouble with two of them, GABARAPL2 antibody (ab15858) and GABARAP antibody (ab1133). I was expected to receive 50 uL of GABARAPL2 antibody and I only received 40 uL. In the case of GABARAP antibody I was expected to recieve 100 uL and I have only received 40 uL. In this case, the recomended dilution for this antibody is 1:500, so I probably won't be able to establish the optimal assay conditions for that antibody with the supplied amount of it. I write you to ask for a solution for this event, because I have paid for a determined amount of antibody and I have received less than the half.
