    IHC image of beta Actin staining in Human normal Colon formalin fixed paraffin embedded tissue section*. The section was pre-treated using pressure cooker heat mediated antigen retrieval with sodium citrate buffer (pH6) for 30mins. The section was incubated with ab8229, 10µg/ml overnight at +4°C. An HRP-conjugated secondary (Ab97100, 1/100 dilution) was used for 1hr at room temperature. The section was counterstained with haematoxylin and mounted with DPX.

    The inset negative control image is secondary-only at 1/500 dilution.

I was not getting any staining in IHC-P using this ab13611, and Envision dual link system HRP secondary antibody. Thanks to your ab97100 free sample, I am now seeing a slight staining pattern at 1:1000 dilution.

Thank you for trying our suggestion of using ab97100 instead of the staining kit. You may want to try increasing the dilution to 1:200 to get stronger staining.

Question

A user has a problem with "Goat polyclonal Secondary Antibody to Rabbit IgG - H&L (HRP) (ab6721)": Antibody solution includes precipitates colored brown.
Even she centrifuged it to eliminate these precipitates as mentioned on the datasheet, she couldn't remove them. So, do you have a suggestion to be able to eliminate them?

In additional, I know that conjugated antibodies are stored at +4 C. But, It is written "store the antibody at -20C" on its datasheet. Could you please check this information?

I have another question: According the user's statement, "Rabbit polyclonal Secondary Antibody to Goat IgG - H&L (HRP) ab97100" was stored at -20C mistakenly. After a overnight, It was realized and stored at +4C. Do you have any experience like this? The user wants to be sure whether this situation affects the antibody's activity /performance.

ab97100; one freeze thaw cycle should not be having any effect on quality so please recommend customer using the antibody as they would like to.

ab6721; please centrifuge out the brown precipitates. You may however need to use the high concentration of antibody than usual but the antibody will be fine. Please let us know if this antibody fails, we will then try to help you with protocol troubleshooting.

Inquiry: Dear Abcam Scientific Support, I'm finding some problems using your primary antibody ab-111976 anti-PAR1.
My protocol is:
8.5% acrylamide gel, 90 minutes of transfer on nitrocellulose or PVDF membrane,
2 hours blocking with 5% milk in TBS (I have also tryed 5% milk in Tween-TBS and 5% BSA in TBS),
primary antibody (diluted 1:500 / 1:1000 in the same blocking buffer) over night at 4°C (I have also tryed 1 hour at RT) and secondary antibody ab97100 Abcam (1:5000 /1:10000) diluted in the same blocking buffer) 1 hour at RT.
After a lot of tests I continue to find high background and my film is all black.
Can you explain me where I'm wrong?

What is the type of sample you are working with? (Tissue / cell lysate, species)
How did you store the antibody?
Did you run a "noprimary control"? did you get a black blot using the secondary only? A "no primary" control would determine if the problem comes from the primary or the secondary.
Could you please confirm you placed an order for this antibodyvia *******?
