      Specific antibodies were absorpted by incubation with Sepharose-bound mouse immunoglobulins. Specific antibodies were eluted by acidic buffer at pH 2.5 followed by neutralisation and dialysis. After repeated binding with immobilized mouse immunoglobulins a minimum of 65% protein bound.
    • Conjugation notes

      The purified polyclonal was conjugated to fluorescein isothiocyanate isomer I followed by gel-filtration and ion-exchange chromatography to clear unbound conjugate.

    Answer

    When antibody is anti-IgG (H+L) then it will react with other immunoglobulin classes (IgM, IgA, IgD, IgE) and subclasses since they all share the same light chains (either kappa or lambda). When antibody is Anti-IgG, F(ab')2 - not specific for IgG- they react with light chains, and therefore also react with other immunoglobulin classes (IgA, IgM, IgD, and IgE) and subclasses sharing the same light chains. Other options anti-IgM compatible secondaries besides ab8517 for Flow Cytometry are: ab6785 Goat polyclonal to Mouse IgG - H&L (FITC) ab60313 Goat polyclonal to Mouse IgG (Chromeo™ 488) ab60316 Goat polyclonal to Mouse IgG - H&L (Chromeo™ 546) ab60318 Goat polyclonal to Mouse IgG (Chromeo™ 642) ab6563 and ab7002 are other options. Please bear in mind that the affinity might be lower for IgG H&L antibodies against IgM, therefore the safest would be to use a secondary against IgM.
