    Cells were cultured in medium supplemented with 50 ng/mL TPA (phorbol, 12 myristate, 13 acetate) for two hours at 37°C immediately prior to harvesting. We recommend aliquoting the extracts into single use fractions and then storing.

  • Background

    HeLa cells are human epithelial cells from a fatal cervical carcinoma. The cell line was derived from cervical cancer cells taken from Henrietta Lacks, in 1951. Horizontal gene transfer from human papillomavirus 18 (HPV18) to human cervical cells created the HeLa genome which is different from either parent genome in various ways including its number of chromosomes. HeLa cells have a modal chromosome number of 82, with 4 copies of chromosome 12 and 3 copies of chromosomes 6, 8, and 17. HeLa cells are adherent cells (they stick to surfaces) and maintain contact inhibition in vitro.

Question

Our customer want to purchase positive control. As your recommendaion, the Jurkat cells treated with anti Fas antibody to induce cleavage is positive control. But I could not find the catalog no in your product. Please let me know the catalog no of this positive control. And customer will use Hela cell so, is it possible to use ab14653 as positive control> I await your reply.

Unfortunately we do not sell the positive control for ab5498, the source of ab5498 has used Jurkat cells treated with a mouse (monoclonal) anti-human Fas (unfortunately not supplied by ourselves, 0.5 µg/mL, 3 hours) or TNF-a to induce cleavage; the laboratory also used caspase-8 cleaved recombinant human BID. Regarding the HeLa lysate, ab14653 is a nuclear lysate of HeLa cells stimulated with TPA for 2hrs and it is therefore a very specific lysate, if the customer wants a whole cell lysate I would not recommend this product and recommend looking at biocompare.com for a product more suitable to his/her request.
