      MCF-7 cells are a well-characterized estrogen receptor (ER) positive control cell line (cells are positive for cytoplasmic estrogen receptors) and therefore are a useful in vitro model of breast cancer to study the role of estrogen in breast cancer. Cells are also positive for cytokeratin and negative for desmin , endothelin , GFAP, neurofilament, vimentin.

      Each lot has been tested for Sp1 activity by using the TransAM™ Sp1 Kit. The signal intensity for Sp1 activity in each lot is compared to the signal intensity obtained with H2O2-post treated extracts from MCF-7 cells. Once the signals are blanked, the ratio between the signals from untreated extracts over treated extracts used at 5 µg/well must be above 4.

    Thank you for contacting us.

    We do not know what effect H2O2 stress will have on expression of BCL9 in MCF7 cells therefore I would recommend perhaps running H2O2 treated MCF7 lysate alongside untreated if possible to determine affect.

    We do have other tissue types that can be used as control signal. OurHeLa Nuclear Lysate, (ab14655:https://www.abcam.com/HeLa-Human-epithelial-carcinoma-cell-line-Nuclear-Lysate-ab14655.html) gave the next best signal after MCF7 with ab37305. This product is in stock with next day delivery.

    Thank you for your enquiry. The MCF-7 nuclear extracts have been collected in Lysis Buffer. The Lysis Buffer consists of 20 mM Hepes pH 7.5, 400 mM NaCl, 20% glycerol, 0.1 mM EDTA, 10 mM NaF, 10 µM Na2MoO4, 1 mM NaVO3, 10 mM PNPP, 10 mM ß-glycerophosphate, 1 mM DTT and protease inhibitor. If the extract was diluted to reach the concentration of 2.5 mg/ml (actual concentration), it is in this lysis buffer. I don't think that if it was diluted it was more than 2 to 3 fold, but I don't have the exact info. The lysate does not contain DNA. I hope this information helps.
