Application Abreviews Notes
WB 1/2000. Detects a band of approximately 23 kDa (predicted molecular weight: 25 kDa).
IHC-P 1/16000. Perform heat mediated antigen retrieval with Tris/EDTA buffer pH 9.0 before commencing with IHC staining protocol.

This antibody is not suitable for human species in IHC application.

IHC-Fr 1/1000.

Perform heat mediated antigen retrieval with 10 mM sodium citrate buffer pH 6.0 + 0.05% Tween-20 before commencing with IHC staining protocol.

Under our IHC-Fr experimental conditions this antibody could not detect Prolactin on rat pituitary gland sample.

    Prolactin expression is generally restricted to the anterior pituitary gland (PMID: 1697858).

Cytoplasmic staining in lactotrophic cells of anterior pituitary gland (PMID: 16468032). The nuclear counter stain is DAPI (blue).

    Secondary antibody only control: Used PBS instead of primary antibody, secondary antibody is Goat Anti-Rabbit IgG H&L (Alexa Fluor® 488) (ab150077) at 1/1000 dilution.

  • Prolactin/PRL was immunoprecipitated from 0.35 mg of mouse pituitary tissue lysate with ab188229 at 1/30 dilution. Western blot was performed from the immunoprecipitate using ab188229 at 1/1000 dilution. VeriBlot for IP Detection Reagent (HRP) (ab131366), was used for detection at 1/10000 dilution.

    Lane 1: Mouse pituitary tissue lysate 10 µg (Input). 

    Lane 2: ab188229 IP in mouse pituitary tissue lysate. 

    Lane 3: Rabbit monoclonal IgG (ab172730) instead of ab188229 in mouse pituitary tissue lysate.

    Blocking and dilution buffer and concentration: 5% NFDM/TBST.

    Exposure time: 1 second.
