  • Product overview

Abcam’s Chemiluminescent Prostaglandin E2 (PGE2) in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Prostaglandin E2 in biological fluids.

A goat anti-mouse IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated-Prostaglandin E2 antibody. After incubation the excess reagents are washed away and a chemiluminescent substrate is added, which causes light emission at approximately 530 nm. The intensity of the emitted light is inversely proportional to the amount of Prostaglandin E2 captured in the plate.

  • Notes

Prostaglandin E (PGE ) is formed in a variety of cells from PGH , which itself is synthesized from

arachidonic acid by the enzyme prostaglandin synthetase. PGE has been shown to have a number

of biological actions, including vasodilation, both anti- and proinflammatory action modulation of sleep/wake cycles, and facilitation of the replication of Human immunodeficiency virus. It elevates cAMP levels and stimulates bone resorption, and has thermoregulatory effects. It has been shown to be a regulator of sodium excretion and renal hemodynamics.

Answer

ab136948 as well as our other PGE2 kits, ab133021, ab133055 and ab136949 are species independent; thus the kit is not specific for any particular species. As long as there enough PGE2 in their rat sample for the kit to detect then the kit should work assuming that the customer has optimized dilution and demonstrated dilutional linearity. ab136948 comes with a goat anti-mouse IgG coated plate so samples containing endogenous mouse IgG’s (such as mouse serum or plasma) may lead to some interference in the assay due to the likelihood for those endogenous mouse IgG’s to bind to the goat anti-mouse IgG antibody on the plate. However, this should not affect rat samples. I have provided a citation below which references using one of our PGE2 kits for rat PGE2 cell lysates.

Answer

1) PGE2 CLIA kit is species independent; thus the kit is not specific for any particular species. However, the kit comes with a goat anti-mouse IgG coated plate so samples containing endogenous mouse IgG’s (such as serum or plasma) may lead to some interference in the assay due to the likelihood for those endogenous mouse IgG’s to bind to the goat anti-mouse IgG antibody on the plate. That being said, for mouse samples not containing endogenous mouse IgG’s and/or for mouse samples that have had the IgG’s removed, the detection antibody in the kit is not species specific so as long as there enough PGE2 in the sample for the kit to detect then the kit should work assuming that you can achieve dilutional linearity.

2) The PGE2 is not from a species source but is a synethically made prostaglandin.

Thank you for contacting us.
The following kits can be used to determine the amount of PGE2 in tissue samples however these kits can not be used for extracting the PGE2.

Answer

Thank you for contacting us.

ab 136948 is species independent for that marker is conserved between species. In addition, the kit comes with a goat anti-mouse IgG plate so you need not worry about endogenous rat IgG’s cause interference either. Therefore, as long as there is enough for the kit to detect then rat should be a compatible species with this kit.
