    Abcam’s Prostaglandin E2 High Sensitivity in vitro competitive ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for the accurate quantitative measurement of Prostaglandin E2 in serum, saliva, urine, tissue culture media and other biological fluids.

    A goat anti-mouse IgG antibody has been precoated onto 96-well plates. Standards or test samples are added to the wells, along with an alkaline phosphatase (AP) conjugated-PGE2 antigen and a monoclonal mouse antibody specific to PGE2. After incubation the excess reagents are washed away. pNpp substrate is added and after a short incubation the enzyme reaction is stopped and the yellow color generated is read at 405 nm. The intensity of the yellow coloration is inversely proportional to the amount of PGE2 captured in the plate.

    Prostaglandin E2 (PGE2) is an extensively studied prostaglandin owing to its predominance in inflammation, cancer, atherosclerosis, autoimmune disease, and sepsis. Oxidation of arachidonic acid by prostaglandin synthases (COX-1 and COX-2) produces prostaglandin H2 (PGH2), which is further metabolized by PGE synthases into its major product, PGE2. PGE2 mediates autocrine and paracrine signaling by binding to G-protein coupled receptors (EP1, EP2, EP3, EP4) on the cell surface, functioning to modulate phospholipase C and adenylate cyclase activity. PGE2 has been of great interest as a therapeutic target, either by modulation of its synthesis by COX inhibitors (NSAIDS) or by modulation of its receptors by downregulation or binding antagonists. PGE2 production in a variety of tissues has been shown to modulate numerous physiological processes including natriuresis in the kidney, smooth muscle elasticity in the vasculature, and the inflammatory response to damaged tissues by monocytes and macrophages.

ab136948 as well as our other PGE2 kits, ab133021, ab133055 and ab136949 are species independent; thus the kit is not specific for any particular species. As long as there enough PGE2 in their rat sample for the kit to detect then the kit should work assuming that the customer has optimized dilution and demonstrated dilutional linearity. ab136948 comes with a goat anti-mouse IgG coated plate so samples containing endogenous mouse IgG’s (such as mouse serum or plasma) may lead to some interference in the assay due to the likelihood for those endogenous mouse IgG’s to bind to the goat anti-mouse IgG antibody on the plate. However, this should not affect rat samples. I have provided a citation below which references using one of our PGE2 kits for rat PGE2 cell lysates.
