We have used our internal sandwich ELISA protocol only to determine which monoclonal antibodies are working together as pairs. Usually any general protocol, like for example the one youfind below, with proper optimization should work with these monoclonal antibodies.


ab10184: Can be paired for Sandwich ELISA with https://www.abcam.com/index.html?datasheet=10185 and https://www.abcam.com/index.html?datasheet=10187 and https://www.abcam.com/index.html?datasheet=24466. For the detection of total PSA, ab10184 can be used as a capture antibody in conjugation with https://www.abcam.com/index.html?datasheet=10185 or with https://www.abcam.com/index.html?datasheet=24466 (HRP-conjugated version). For the detection of free PSA, ab10184 can be used as a capture antibody in conjugation with https://www.abcam.com/index.html?datasheet=101887.

ab10185: Can be paired for ELISA with https://www.abcam.com/index.html?datasheet=10184, and https://www.abcam.com/index.html?datasheet=10187.

ab10187: Can be paired for Sandwich ELISA with https://www.abcam.com/index.html?datasheet=10184 and https://www.abcam.com/index.html?datasheet=10185 and https://www.abcam.com/index.html?datasheet=24466. General Sandwich ELISA protocol

1. Add coating antibody at 5 μg/ml concentration in phosphate buffered saline (PBS, pH 7.4) on the 96-well EIA plate, incubate overnight at + 4 °C

2. Wash 2-3 times with PBST, 200 μl/well, for one minute at RT

3. Add two-fold serial dilution of the antigen in PBS-AT, 100 μl/well, incubate for 1 hour at + 37 oC.

4. Wash 3 times with PBST, 200 μl/ well, for one minute at RT.

5. Add HRP-conjugated MAb at 0.2-1 μg/ml concentration in PBST, 100 μl/well, incubate for 1 hour at + 37 oC.

6. Wash 2-5 times, 200 μl PBST per well for one minute at RT.

7. Add 100 μl of TMB substrate per well and incubate for 15-30 min at RT.

8. Stop the reaction with sulphuric acid, incubate for 5 minutes

9. Read the absorbance at 450 nm.

This is a general protocol for sandwich ELISA immunoassay. Optimal antibody and antigen working concentrations as well as incubation times and buffers should be determined separately by the user.

I have reviewed the details of this experiment and have the following feedback:

1. ab119154 Prostate Specific Antigen protein is full length human protein, so should be detected by both ab77310 and ab102885.

2. The ab102885 Easylink conjugation kit would be suitable for conjugating FITC to ab10187.

3. In theory, the experiment the customer has described should work. However, these antibodies have not been tested in this type of application, an I am sorry we would not be able to guarantee them for this.

4. Also, as discussed in the previous email, for sandwich ELISa type experiments, we would suggest to use a matched pair. ab10187 has been tested as a matched pair withthe following:
