    Bonjour Je vous remercie grandement de votre réponse. Pour répondre à vos questions concernant mon protocole de Western Blot :- J’ai bloqué la membrane avec du lait 5% dilué dans du PBS-Tween pendant 45 minutes- L’anticorps anti-PSA ab2218 a été dilué au 1/100 dans du PBS-Tween 1.5% lait puis incubé overnight à 4°C- J’ai utilisé un anticorps Goat anti Mouse HRP utilisé à 1/10000 dans du PBS-Tween 1.5% lait puis incubé 1h à RTL’anticorps a été testé sur les cellules C4-2 humaines (doit être exprimé), un lysat de prostate primate, des lysats de peau de souris (ne doit pas être exprimé normalement). Nous voyons juste les bandes aspécifiques à 25Kda chez la souris qui n’exprime pas le PSANous avons commencé à regarder vos autres AC anti-PSA, mais peu pourraient nous aller, puisque nous cherchons un anti-PSA qui crosse aussi avec le cynomolgus.Pour le kit ELISA, je reviendrais vers vous après discussion avec ma supérieure car c’est délicat d’acheter encore un kit sans être sure de son efficacité pour le Cynomolgus. Voilà j’espère avoir répondu correctement à vos questions, n’hésitez à me revenir vers moi si vous en avez l’utilité.

    Merci pour votre réponse.
    Pour le ab2218, je vous conseille de tester cet anticorps aussi sur des membranes bloquées avec de la BSA. Effectivement, de changer le tampon de blocage peux influencer significativement le résultat. Ceci est illustré par exemple dans un image Western blot sur la fiche technique de ab9385.     Si ce changement de tampon n'améliore pas non plus les résultats, nous allons vous remplacer aussi cet anticorps. Je suis désolée que je ne trouve pas des autres solutions.
    Pour ce qui est des autres anticorps, je pense qu'il y ait des fortes chances pour que certains cross réagissent. Les séquences des protéines respectives humaines et de macaque sont assez proches. http://www.ebi.ac.uk/Tools/services/web_clustalw2/toolresult.ebi?tool=clustalw2&jobId=clustalw2-I20120302-114412-0654-17499195-pg Nous pourrions donc vous faire des offres de test comme pour le kit www.abcam.com/collaborationdiscount.

    Bonjour

    Je me permets de vous recontacter suite au remplacement de mon anticorps anti-PSA par l’Ac anti-PSA ab2218.
    Cette fois-ci je vois bien une bande mais elle est totalement aspécifique puisque je retrouve le PSA chez les organes de souris (alors que la souris n’exprime normalement pas le PSA ??) mais par contre pas dans les C4-2 humaines qui elles sont censées l’exprimer.

    Dans tous les cas, la bande obtenue même pour la souris est aspécifique puisque pas à taille recherchée (=34kDa).

    Je vous mets ci-dessous mon Western blot.

    Je ne comprends pas vraiment ces résultats. L’anticorps secondaire a déjà été testé avec d’autres Anticorps primaires (comme la tubuline) et il n’y a aucun souci.

    Auriez-vous quelque chose à me recommander ?

    J’ai vu qu’il y avait une promotion en ce moment chez vous et je souhaiterais tester d’autres anticorps PSA.

    De plus, auriez-vous un kit ELISA PSA qui serait capable de matcher avec le Cynomolgus macaca (ou fascicularis), car je ne vois pas les spécificités d’espèces sur votre site pour ce type de produit.

    Merci pour votre émail. Je suis très désolée d'entendre que le nouveau anticorps ne fonctionne pas non plus comme attendu.
    Pour l'instant nous n'avons pas eu des problèmes avec cet anticorps non plus et je ne sais pas donc ce qui peux être à l'origine du problème.
    Est-ce que vous avez testé l'anticorps secondaire aussi sur les lysats de peau? Effectivement, je peux conseiller ceci pour exclure que il ne reconnait pas la chaîne légère des immunoglobulines présents dans la peau de souris. Comme vous, je ne pense pas que cette bande aux alentours de 25kD soit spécifique.
    Pour ce qui est de l'absence de la bande spécifique, j'aimera bien vous poser quelques questions additionnelles:
    1.) A quel dilution est-ce que vous avez utilisé le ab2218?
    2.) Quel tampon de blocage est-ce que vous avez utilisé?
    3.) Dans quel tampon et pour combien de temps est-ce que vous avez fait les incubations des anticorps?
    Je me réjouis de votre réponse afin de voir comment nous pouvons procéder pour cet anticorps.
    Effectivement nous avons des promotions actuellement. Je vais volontiers vous assister pour sélectionner des autres anticorps si vous souhaitez.
    Nous avons des kits ELISA qui sont testés sur la protéine humaine seulement. Mais je peux vous faire une offre pour les tester sur du Cynomolgus. Les deux kits que nous avons sont le ab108656 et le ab113327.

    Thank you for your email. I can't tell you the name of the originator of ab9537 as that information is proprietary. I double-checked ab403 and ab2218, and both of these do not contain BSA or other proteins in the storage buffer. Ab403 consists of preservative: 0.1% Sodium Azide, and constituents: sodium chloride, citrate / phosphate. pH 6.0. Ab2218 consists of preservative: 15mM sodium azide, and constituents: PBS. pH 7.4.
