Thank you for your swift reply. The samples are centrifuged and the supernatant is 0.2µm-filtered. However, in some cases, centrifugation does not produce any supernatant and the substrate is too viscous for filtration. Even after dilution and above treatment there might remain humic acid in the sample and turbidity that can influence results. Experiences with enzyme activities in slurries,manure, compost are very limited and results not standardized. I guess activities are in the range of ng-µg peptides/g dm per hr or ng-µg peptides/ml per hr.
Indeed I have another question: while the expected activities seem to match with your kits, it would be useful to know how "wide" the linear part of your standard curve is.
I suppose that your lower limit is somewhere at 2ng/mlxhr. What is your upper limit in the same sample?
The reason is that we assume that enzyme activities, in manure for example, differ considerably depending on the samples. Preparation of time consuming dilution series could be prevented.
I suppose we are going to test some kits available on the market. What are the chances that we get "sample" assay kits to run some tests instead of buying 100-500 test kits with your company?

Thank you again for your patience while I have been in touch with the lab regarding these kits.
The kit ab111750 should not be used with turbid or non-clear samples, but my contact at the lab did say that ab112152 or ab112153 might be ok to use. We have not tested them in this way so we can't say for sure. For all of these kits, the upper limit of the linear standard curve has not been fully determined at this time. Ab112152 and ab112153 have been tested with up to 10 ug/mL of trypsin without any problems.
