  • Function

    Phosphodiesterase (PDE) that has higher activity toward cAMP than cGMP, as substrate. Plays a role in cell proliferation, is able to induce cell motility and acts as a negative regulator of NME1.

    Ubiquitously expressed. Seems to be overexpressed in aggressive sarcoma subtypes, such as leiomyosarcomas and malignant fibrous histiocytomas (MFH) as well as in the less malignant liposarcomas.

  • HepG2 (human liver hepatocellular carcinoma cell line) cells stained for PRUNE (green) using ab237518 at 1/135 dilution in ICC/IF, followed by Alexa Fluor 488-congugated Goat Anti-Rabbit IgG (H+L) secondary antibody.

    The cells were fixed in 4% formaldehyde, permeabilized tissue using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. Counterstained with DAPI.

  • Paraffin-embedded human lung tissue stained for PRUNE using ab237518 at 1/300 dilution in immunohistochemical analysis.

    After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30min at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized tissue using an HRP conjugated SP system.

  • PRUNE was immunoprecipitated from 500 μg HepG2 (human liver hepatocellular carcinoma cell line) whole cell lysate.  For western blotting, a HRP-conjugated Protein G antibody was used as the secondary antibody (1/2000).
