Involvement in disease
Defects in PTCH2 are a cause of medulloblastoma (MDB) [MIM:155255]. MDB is a malignant, invasive embryonal tumor of the cerebellum with a preferential manifestation in children. Although the majority of medulloblastomas occur sporadically, some manifest within familial cancer syndromes such as Turcot syndrome and basal cell nevus syndrome (Gorlin syndrome).
Defects in PTCH2 are a cause of sporadic basal cell carcinoma (BCC) [MIM:605462]. -

Paraffin-embedded human brain tissue stained for PTCH2 using ab238338 at 1/200 dilution in immunohistochemical analysis.
After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum 30 minutes at room temperature. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.

HEK-293 (Human epithelial cell line from embryonic kidney) cells stained for PTCH2 (green) using ab238338 at 1/66 dilution in ICC/IF. Secondary antibody is an Alexa Fluor® 488-conjugated Goat Anti-Rabbit IgG (H+L).
The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal goat serum. The cells were then incubated with the antibody overnight at 4°C.
