    Phosphorylase is an important allosteric enzyme in carbohydrate metabolism. Enzymes from different sources differ in their regulatory mechanisms and in their natural substrates. However, all known phosphorylases share catalytic and structural properties.
  • Involvement in disease

    Defects in PYGL are the cause of glycogen storage disease type 6 (GSD6) [MIM:232700]. A metabolic disorder characterized by mild to moderate hypoglycemia, mild ketosis, growth retardation, and prominent hepatomegaly. Heart and skeletal muscle are not affected.

The PYGL antibody ab103419 is raised against the full-length protein, amino acids 1-847. The sequence is described in the UniProt entry, accession P06737. Here is a link to the entry. http://www.uniprot.org/uniprot/P06737.

It is a polyclonal antibody so it is expected to recognize multiple epitopes on PYGL. Since human PYGL and PYGM are identical at 81% of the amino acids of PYGL, it is likely the antibody will cross-react with PYGM. But we have not tried to confirm this experimentally.

The PYGM antibody ab88078 is raised against an immunogen consisting of amino acids 734-843, http://www.uniprot.org/uniprot/P11217 . This is a monoclonal antibody, so it recognizes only one epitope but we do not know where the epitope is located on the amino acid sequence of the immunogen. The immunogen shares 73% identity with the human PYGL amino acid sequence. So, there is some possibility of cross-reaction of the antibody with PYGL. This has not been tested experimentally.
