    • Function

      RNA-binding protein that plays a central role in myelinization (PubMed:16641098). Binds to the 5'-NACUAAY-N(1,20)-UAAY-3' RNA core sequence. Regulates target mRNA stability (PubMed:23630077). In addition, acts by regulating pre-mRNA splicing, mRNA export and protein translation. Required to protect and promote stability of mRNAs such as MBP and CDKN1B. Regulator of oligodendrocyte differentiation and maturation in the brain that may play a role in myelin and oligodendrocyte dysfunction in schizophrenia (PubMed:16641098). Participates in mRNA transport by regulating the nuclear export of MBP mRNA. Also involved in regulation of mRNA splicing of MAG pre-mRNA. Acts as a translational repressor.

    • Immunocytochemistry/ Immunofluorescence analysis of Neuro-2a (Mouse neuroblastoma neuroblast) cells labeling QKI with Purified ab126742 at 1:100 dilution (5.6μg/ml). Cells were fixed in 4% Paraformaldehyde and permeabilized with 0.1% tritonX-100. Ab150077 Goat anti rabbit IgG(Alexa Fluor® 488) was used as the secondary antibody at 1:1000 dilution. DAPI nuclear counterstain. PBS instead of the primary antibody was used as the secondary antibody only control.

    In regards to the immunogen sequence which you have requested. Ab78130 is specific for QK1 isoform 1, known also asHQK-5 or QKI-5. Ab78518 is directed against isoform 4 otherwise known asHQK-7.

    Though I have been unable to find the exact immunogen used to create these products, the HQK-7isoform differs from the canonical (HQK-5, sequence as follows:
    http://www.uniprot.org/blast/?about=Q96PU8[312-341]: GAVATKVRRHDMRVHPYQRIVTADRAATGN→EWIEMPVMPDISAH Therefore the immunogen specific to this product should be within this region.

    I have passed your enquiry regarding the immunogen sequence for ab78518 on to the lab. I will forward you any information I receive.

    The concentration of these 3 antibodies has not been determined. ab78518 and ab78130is not determined. These are both whole antiserum products and not purified antibodies. However, estimated concentrationof specific primary antibody for whole serum products is about 0.5mg/ml. And ab126742is a tissue culture supernatant product. This is also not a purified antibody and the specific antibody concentration in this sort of productis typically 0.05mg/ml.
