    Use at an assay dependent concentration. Detects a band of approximately 22 kDa (predicted molecular weight: 24 kDa). By Western blot, this antibody detects an ~22 kDa protein representing Rab 11 in CV1, PC3, and HeLa cell extract. Another less prominent, unknown band is also detected by this antibody at ~35 kDa.

    The cells I am using are HEK293 cells, so they are human. The protein concentration is unknown (I haven’t quantitated it), however, the same sample was used for Rab5 (ab18211, a marker for early endosomes) and beta-actin (ab8227), and those antibodies worked well. I am assuming there would be realitvely similar quantities of late (Rab7) and recycling (Rab11) endosomes to early endosomes (Rab5), but perhaps I am mistaken?

    The lysis buffer I am currently using is M-PER Mammalian Protein Extract Reagent from Thermo Scientific, so naturally the recipe is proprietary.

    I have been leaving the primary antibody on for 1hr, at room temperature, on a rocking platform

    I was originally blocking in 3% BSA overnight, but switched to 1% BSA for 1hr and still am seeing no bands

    Yes, the secondary antibody I am using has worked with Rab5 and beta-actin
    Any suggestions would be very useful!

    Answer

    Thank you for your reply. I would recommend keeping the blocking to one hour and incubating the primary antibody overnight at 4 degrees. I would suggest doing this at 1/300 for ab74906 and at 1/1000 for ab3612. Please let me know if this does not improve your results.

    Inquiry: Dear Sir or Madam, I am currently looking for an anti-human Rab 11 antibody for immunocytochemistry to perform localization studies. Unfortunately, we have had negative experience with antibodies that were not specific or not sensitive enough for confocal imaging of Rab11. Are there any means to test the antibody before buying the lot? We would be very grateful if it was possible to obtain a free tester sample sufficient for staining 2 - 3 coverslips of the anti-Rab 11 rabbit polyclonal antibody (ab3612). As our lab is buying quite frequently at abcam, we could arrange including the sample into next package to avoid extra shipping payments. I will greatly appreciate receiving any information on anti-Rab 11 antibodies (advise, new products, etc.) that you can provide. Yours sincerely,

    Thank you for the further information. It is difficult to select alternatives that are the most 'specific' as we have not tested these antibodies side by side in the same sample application or species. Reviewing the datasheets I think that ab74906 anti-Rab7 antibody and ab3612 anti-Rabb11 antibody may be preferable but please do check the datasheets to confirm whether you agree. Please bear in mind that ab74906 (rabbit polyclonal to Rab7) is not tested in ICC, and as it shows a weak band in WB the staining intensity may be lower than other antibodies. In addition, ab3612 (rabbit polyclonal to Rab11) is only predicted to work in mouse samples, and may not give positive results. As the Rab7 and Rab11 antibodies will be used in previously untested species/applications, would it be possible to submit an Abreview with your results? It doesn't matter if it is positive or negative, but the information will be really useful for other researchers that may be interested in using these products in a similar manner. I will be able to provide ab78970 and ab87802. Could you also submit Abreview information for these products too? Any other information you be able to share via our Abreview system would be extremely useful. Looking forward to your reply.

    Thank you for contacting us. Yes, a suggested starting dilution for the primary antibody would be 1/200. And I would suggest keeping the secondary antibody dilution constant while titrating the primary antibody. Once you know the optimal dilution for the primary you could try a higher and a lower dilution of the secondary to see if any other dilution might give even better results.
