Properties

Applications

Our Abpromise guarantee covers the use of ab3612 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use at an assay dependent concentration. PubMed: 18384686
WB Use at an assay dependent concentration. Detects a band of approximately 22 kDa (predicted molecular weight: 24 kDa).Can be blocked with Human Rab11 peptide (ab41779). By Western blot, this antibody detects an ~22 kDa protein representing Rab 11 in CV1, PC3, and HeLa cell extract. Another less prominent, unknown band is also detected by this antibody at ~35 kDa.
IP Use at an assay dependent concentration. PubMed: 17686995

Target

Images

  • Immunofluorescence analysis of HeLa cells, staining Rab11 (red) with ab3612.
Cells were fixed with paraformaldehyde for 10 minutes, blocked in PBS with 5% normal goat serum and permeabilized with 0.1% Triton X-100. Samples were incubated with primary antibody (1/200). An AlexaFluor®594-conjugated anti-rabbit IgG (1/1000) was used as the secondary antibody.

  • ab3612 staining HEK 293 cells by ICC/IF.  Cells were PFA fixed and permeabilized in 0.012% saponin prior to blocking in 1% BSA for 30 minutes at 37°C.  The primary antibody was diluted 1/200 and incubated with the sample for 2 hours at 37°C. A biotinylated goat anti-rabbit antibody, diluted 1/200, was used as the secondary.

This image depicts ab3612 recognizing endogenous Rab11 (red). Rab11 was used as a recycling endosome marker.

  • ab3612 used on PC3 cell extract.

  • ab3612 staining Rab11 in Canine kidney cells by Immunocytochemistry/ Immunofluorescence.Cells were fixed in paraformaldehyde, permeabilized using 0.5% saponin, then incubated with ab3612 at a 1/200 dilution for 1.5 hours at 20°C. The secondary used was an Alexa-Fluor 568 conjugated donkey anti-rabbit polyclonal, used at a 1/350 dilution.

Answer

Thank you for contacting us. Because we carry over 90,000 products, it isn't feasible for us to keep small sample sizes of our products.

We are happy to reassure our customers that all of our products are covered by our Abpromise, which guarantees that the product will work in the applications and species specified on the datasheet, or we will offer a replacement, credit, or refund within 6 months of purchase.

If the product is to be used in an untested species or application, you may be eligible for our testing discount program if the antibody has not yet been purchased. Please contact our Scientific Support team by replying to this email prior to purchase for more information.

As shown on the product datasheet, the Rabbit polyclonal anti-Rab11 ab3612 (https://www.abcam.com/ab3612) is guaranteed to work in ICC/IF, Western Blot and on Human, Dog, Non Human Primates samples.
The other anti-Rab11 we have in our catalogue is a Rat monoclonal, reference ab95375 (https://www.abcam.com/ab95375). Like ab3612 it has been tested in ICC/IF and Western Blot but only on Mouse samples.

I hope this information is helpful. Please do not hesitate to contact us again with any other questions.

Question
Answer

Thank you for the further information. It is difficult to select alternatives that are the most 'specific' as we have not tested these antibodies side by side in the same sample application or species. Reviewing the datasheets I think that ab74906 anti-Rab7 antibody and ab3612 anti-Rabb11 antibody may be preferable but please do check the datasheets to confirm whether you agree. Please bear in mind that ab74906 (rabbit polyclonal to Rab7) is not tested in ICC, and as it shows a weak band in WB the staining intensity may be lower than other antibodies. In addition, ab3612 (rabbit polyclonal to Rab11) is only predicted to work in mouse samples, and may not give positive results. As the Rab7 and Rab11 antibodies will be used in previously untested species/applications, would it be possible to submit an Abreview with your results? It doesn't matter if it is positive or negative, but the information will be really useful for other researchers that may be interested in using these products in a similar manner. I will be able to provide ab78970 and ab87802. Could you also submit Abreview information for these products too? Any other information you be able to share via our Abreview system would be extremely useful. Looking forward to your reply.

Answer

Thank you for contacting us. Yes, a suggested starting dilution for the primary antibody would be 1/200. And I would suggest keeping the secondary antibody dilution constant while titrating the primary antibody. Once you know the optimal dilution for the primary you could try a higher and a lower dilution of the secondary to see if any other dilution might give even better results. But it is often advisable to change only one parameter the time. I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information.
