Ran Activation Assay Kit (ab211172) utilizes RanBP1 Agarose beads to selectively isolate and pull-down the active form of Ran from purified samples or endogenous lysates. Subsequently, the precipitated GTP-Ran is detected by western blot analysis using an anti-Ran antibody, which reacts with the human, mouse, rat, dog and chicken protein.
Features: 1) non radioactive assay format; 2) fast results: 1 hour assay plus electrophoresis/blotting time; 3) includes Cdc42 positive control; 4) pink colored agarose beads for easy identification during washing and aspiration steps.
Small GTP-binding proteins (or GTPases) are a family of proteins that serve as molecular regulators in signaling transduction pathways.
Ran (Ras-related nuclear protein), a 26 kDa protein of the Ras superfamily, is involved in a variety of biological response pathways that include chromatin condensation, cell cycle progression, and translocation of RNA/proteins through the nuclear pore complex. Ran acts a negative regulator of the kinase activity of CRK1 and VRK2. Like other small GTPases, Ran regulates molecular events by cycling between an inactive GDP-bound form and an active GTP-bound form. In its active (GTP-bound) state, Ran binds specifically to RanBP1 to control downstream signaling cascades.

FunctionGTP-binding protein involved in nucleocytoplasmic transport. Required for the import of protein into the nucleus and also for RNA export. Involved in chromatin condensation and control of cell cycle (By similarity). The complex with BIRC5/ survivin plays a role in mitotic spindle formation by serving as a physical scaffold to help deliver the RAN effector molecule TPX2 to microtubules.
