Properties

Applications

Our Abpromise guarantee covers the use of ab31120 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
IHC-P Use at an assay dependent concentration. Perform heat mediated antigen retrieval before commencing with IHC staining protocol.

Ready to use for 30 min at RT. Staining of formalin-fixed tissues requires boiling tissue sections in 10mM citrate buffer, pH 6.0 for 10 min followed by cooling at RT for 20 min

ICC/IF 1/1.

Please refer to ICC/IF image below for further details.

WB 1/10 - 1/30. Detects a band of approximately 23 kDa (predicted molecular weight: 26 kDa).

Please refer to WB image below for further details.

Target

  • Function
GTP-binding protein involved in nucleocytoplasmic transport. Required for the import of protein into the nucleus and also for RNA export. Involved in chromatin condensation and control of cell cycle (By similarity). The complex with BIRC5/ survivin plays a role in mitotic spindle formation by serving as a physical scaffold to help deliver the RAN effector molecule TPX2 to microtubules.
Enhances AR-mediated transactivation. Transactivation decreases as the poly-Gln length within AR increases.
  • Tissue specificity
Expressed in a variety of tissues.
  • Sequence similarities
Belongs to the small GTPase superfamily. Ran family.
  • Post-translational
    modifications
    The N-terminus is blocked.
  • Cellular localization
Nucleus. Cytoplasm. Melanosome. Becomes dispersed throughout the cytoplasm during mitosis. Identified by mass spectrometry in melanosome fractions from stage I to stage IV.

Images

  • Human breast stained with ab31120 for 30 minutes at room temperature.
  • ab31120 staining Ran in HeLa and mouse NIH-3T3 cells, by immunofluorescence. This antibody interacts specifically well with the spindle pole-associated fraction of Ran in mitosis (see image above) and may be extremely useful in studies of mitotic functions of Ran.

    Experimental conditions:

Ab31120 staining Ran in Hela and NIH-3T3 cells, by western blotting. This antibody worked very well with both protein extracts, although some non-specific bands were occasionally seen using concentrated  antibody.

    Optimal antibody dilution: 1/30

WB was performed following a standard protocol: blocking: 30 min; primary antibody: 1 hr; secondary antibody: 30 min; and developed with ECL Plus (Amersham).
