Answer

The freezing point of a 20% glycerol solution is about -5C, so freezing at -20C is expected. If you have aliquotted the antibody into small volumes for single experiments, then this will not be a problem. Freezing and thawing can be damaging but not always. It usually depends on the number of freeze-thaw cycles, so the reason for aliquotting into small volumes is to minimize the number of cycles.

If you want to keep the antibody from freezing at -20C, you can add 60ul of glycerol to the 100ul of antibody you received, which will bring the percentage of glycerol to 50%. The freezing point of a 50% solution is -22C.
