    IGF1 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of IGF1 protein in rat serum, plasma and tissue extract.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    Insulin-like growth factor I (IGF1) is a 70-amino acid hormone that is proteolytically processed from a larger precursor protein.  IGF1 is structurally similar to insulin and has growth promoting effects but has higher growth-promoting activity. IGF1 is thought to stimulate glucose transport in bone-derived osteoblastic cells and is effective at much lower concentrations than insulin. May play a role in synapse maturation. Ca2+-dependent exocytosis of IGF1 is required for sensory perception of smell in the olfactory bulb. IGF1 is a ligand for IGF1R. Binds to the alpha subunit of IGF1R, leading to the activation of the intrinsic tyrosine kinase activity which autophosphorylates tyrosine residues in the beta subunit thus initiatiating a cascade of down-stream signaling events leading to activation of the PI3K-AKT/PKB and the Ras-MAPK pathways.

  • Function

    The insulin-like growth factors, isolated from plasma, are structurally and functionally related to insulin but have a much higher growth-promoting activity. May be a physiological regulator of [1-14C]-2-deoxy-D-glucose (2DG) transport and glycogen synthesis in osteoblasts. Stimulates glucose transport in rat bone-derived osteoblastic (PyMS) cells and is effective at much lower concentrations than insulin, not only regarding glycogen and DNA synthesis but also with regard to enhancing glucose uptake.

    Defects in IGF1 are the cause of insulin-like growth factor I deficiency (IGF1 deficiency) [MIM:608747]. IGF1 deficiency is an autosomal recessive disorder characterized by growth retardation, sensorineural deafness and mental retardation.

  • The IGF1 standard curve was prepared as described in Section 10.

  • The concentrations of IGF1 were measured in duplicates, interpolated from the IGF1 standard curves and corrected for sample dilution. Undiluted samples are as follows: serum 1:200, plasma (citrate) 1:100, and plasma (heparin) 1:200. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGF1 concentration was determined to be 295.70 ng/mL in serum, 149.61 ng/mL in plasma (citrate) and 367.12 ng/mL in plasma (heparin).

  • The concentrations of IGF1 were measured in duplicate and interpolated from the IGF1 standard curve and corrected for sample dilution. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean IGF1 concentration was determined to be 652.52 pg/mL.
