    Abcam’s IL-17A Rat in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for accurate quantitative measurement of Rat IL-17 (Interleukin-17)concentrations in Cell culture supernatant, serum and plasma (EDTA, citrate, heparin).


    IL-17A specific antibodies have been precoated onto 96-well plates. Standards and test samples are added to the wells along with a biotin-conjugated IL-17A detection antibody then incubated at room temperature. Following washing, a Streptavidin-HRP conjugate is added to each well, incubated at room temperature and washed. TMB is added and then catalyzed by HRP to produce a blue color product that changes into yellow after addition of acidic stop solution. The density of yellow coloration is directly proportional to the amount of IL-17A captured on the plate.

Thank you for your patience in this matter. There will be a big difference in the height of the OD values measured before and after addition of the stop solution, so you should take your readings AFTER the stop solution.

I would also recommend setting the shaking speed at 400 rpm – we have seen that shaking the plate during the incubation time increases the OD values significantly.

Samples can vary from mouse to mouse because all mice (even within the same stains) to do not always express proteins at the same levels. Therefore a titration may be necessary.

However, if instructions in our protocol have been followed, samples have been diluted 1:2 (50ul sample + 50ul Sample Diluent), the concentration read from the standard curve must be multiplied by the dilution factor (X2).

The Anti-PPAR gamma antibody (ab27649) is provided already reconstituted in liquid form in a PBS buffer (pH 7.4) with 1% BSA and 0.02% sodium azide supplemented. The customer therefore does not need to reconstitute it. I am sorry but I do not think I fully understand the question in regards to how many tested can be performed with this antibody. We would advise trying the antibody at a dilution of 1/1000 for Western blotting and 1/200 for IHC. However these are only guidelines and further optimisation of the protocol may be required to obtain the best results.

We do not have set expiration dates for our products. Most antibodies are stable and can last for anywhere from a few months to several years if stored properly, so we strongly recommend that you follow the storage instructions on the datasheet for the antibody and kit. These conditions will vary among our products, therefore, it is important to verify the storage conditions for each of our products when you receive them. We guarantee all of our products to work for at least 120 days from the date of purchase when stored correctly.
