    KIM-1 in vitro SimpleStep ELISA® (Enzyme-Linked Immunosorbent Assay) kit is designed for the quantitative measurement of KIM-1 protein in Rat plasma, serum, urine, and cell culture supernatant.


    The SimpleStep ELISA® employs an affinity tag labeled capture antibody and a reporter conjugated detector antibody which immunocapture the sample analyte in solution. This entire complex (capture antibody/analyte/detector antibody) is in turn immobilized via immunoaffinity of an anti-tag antibody coating the well. To perform the assay, samples or standards are added to the wells, followed by the antibody mix. After incubation, the wells are washed to remove unbound material. TMB substrate is added and during incubation is catalyzed by HRP, generating blue coloration. This reaction is then stopped by addition of Stop Solution completing any color change from blue to yellow. Signal is generated proportionally to the amount of bound analyte and the intensity is measured at 450 nm. Optionally, instead of the endpoint reading, development of TMB can be recorded kinetically at 600 nm.


    KIM-1, also known as Kidney Injury Molecule 1 and Hepatitis A virus cellular receptor 1 homolog (HAVcr-1), or T-cell immunoglobulin and mucin domain 1 (TIM-1) is a 283- amino acid protein that contains an N-terminal immunoglobulin-like domain and is encoded by the HAVCR1 gene. KIM-1 is a single-pass type 1 membrane protein has been identified as a receptor for hepatitis A virus. The TIM gene family participates in host immune response. Urinary KIM-1 levels are elevated in nephropathy and is thought to be a biomarker for renal damage.

Undiluted samples are as follows: serum 25%, plasma (citrate) 25%, plasma (EDTA) 25%, plasma (heparin) 12.5%, and urine 25%. The interpolated dilution factor corrected values are plotted (mean +/- SD, n=2). The mean KIM-1 concentration was determined to be 1,435 pg/mL in serum, 1,332 pg/mL in plasma (citrate), 1,444 pg/mL in plasma (EDTA), 1,726 pg/mL in plasma (heparin), and 2,005 pg/mL in urine.
