    Abcam’s TGF beta 1 Rat in vitro ELISA (Enzyme-Linked Immunosorbent Assay) kit is designed for accurate quantitative measurement of Rat TGF beta 1 concentrations in Cell culture supernatant, serum and plasma (EDTA and heparin).

    Rat TGF beta 1 specific antibodies have been precoated onto 96-well plates. Standards and test samples are added to the wells and then incubated at room temperature. After washing, a Biotin-conjugated anti-Rat TGF beta 1 detection antibody is added then incubated at room temperature. Following washing Streptavidin-HRP conjugate is added to each well, incubated at room temperature then again washed. TMB is added and then catalyzed by HRP to produce a blue color product that changes into yellow after adding acidic stop solution. The density of yellow coloration is directly proportional to the amount of Rat TGF beta 1 captured on the plate.

  • Function

    Multifunctional protein that controls proliferation, differentiation and other functions in many cell types. Many cells synthesize TGFB1 and have specific receptors for it. It positively and negatively regulates many other growth factors. It plays an important role in bone remodeling as it is a potent stimulator of osteoblastic bone formation, causing chemotaxis, proliferation and differentiation in committed osteoblasts.
  • Tissue specificity

    Highly expressed in bone. Abundantly expressed in articular cartilage and chondrocytes and is increased in osteoarthritis (OA). Co-localizes with ASPN in chondrocytes within OA lesions of articular cartilage.
  • Involvement in disease

    Defects in TGFB1 are the cause of Camurati-Engelmann disease (CE) [MIM:131300]; also known as progressive diaphyseal dysplasia 1 (DPD1). CE is an autosomal dominant disorder characterized by hyperostosis and sclerosis of the diaphyses of long bones. The disease typically presents in early childhood with pain, muscular weakness and waddling gait, and in some cases other features such as exophthalmos, facial paralysis, hearing difficulties and loss of vision.

      Here is the correct protocol section:

      Prepare your serum and plasma samples before starting the test
      procedure. Dilute serum and plasma samples with Assay Buffer (1x)
      according to the following scheme:
      - 20 μl sample + 920 μl Assay Buffer (1x)
      - Add 30 μl 1N HCl to 940 μl prediluted sample, mix and incubate for
      1 hour at room temperature.
      - Neutralize by addition of 30 μl 1N NaOH.
      Prepare your cell culture supernatant samples before starting the test
      procedure. Dilute cell culture supernatant samples with Assay Buffer
      (1x) according to the following scheme:
      - 20 μl sample + 180 μl Assay Buffer (1x)
      - Add 20 μl 1N HCl to 200 μl prediluted sample, mix and incubate for
      1 hour at room temperature.
      - Neutralize by addition of 20 μl 1N NaOH.

      The table is correct.

      We have not tested bronchoalveolar lavage fluid with ab119558, so we do not have a recommended dilution for this sample type. If you perform a literatire search to determine the expected levels of TGF beta 1 in this fluid compared to serum, plasma, or cell culture supernatant, then you can use the expected level as a guide to what dilution to use.

      We always recommend testing multiple different dilutions of your sample to ensure that the readings fall within the range of the standard curve.

      Our TGF beta 1 Rat ELISA Kit will detect the inactive ( which get activated during the sample preparation (HCL and NaOH)) and active form.

      It will not detect the LAP complex.
