Properties

Applications

Our Abpromise guarantee covers the use of ab22492 in the following tested applications.

The application notes include recommended starting dilutions; optimal dilutions/concentrations should be determined by the end user.

Application Abreviews Notes
ICC/IF Use at an assay dependent concentration.
IHC-Fr Use at an assay dependent concentration. Fix with acetone.
IHC-P Use a concentration of 10 µg/ml.

Target

  • Relevance
RECA-1 reacts with rat endothelial cell antigen and showed to be reactive with all vascular endothelium in three tested rat strains of different MHC-haplotype; Lewis (TR-11), BN (RT-1n) and OA (RT-1u). RECA-1 recognises a cell surface antigen. The antibody has been successfully applied in staining of viable endothelial cells in vitro, and of vascular endothelium in vivo. No reactivity of RECA-1 was seen with other cells e.g. fibroblasts, leukocytes and non endothelial stromal cells nor with other various tested species other than rat e.g. mouse, rabbit, sheep, goat and human. RECA-1 is a promising antibody for rat endothelial cell studies, and in particular for further defining nature and function of endothelial cell-specific antigens.

Question

Inquiry: Dear abcam Your datasheet for ab22492 states that it has been updated on 6 september 2012. It still states that ab22492 is unsuitable for IHC-P and only for IHC-Fr. However, according to what I have read below, your answer to the customer's question is that ab22492 can be used for both IHC-P and IHC-Fr. Hence I am confused as to which I should believe - the updated datasheet for ab22492 or your reply to this customer's inquiry dated 31 May 2012. Pls clarify soon so I could buy the correct antibody from you.
Inquiry: Hi, I have a question about 'specific references' for your antibodies: I was looking through the specific references for the antibody ab22492 (anti-RECA1), and a paper by Lee et al is referenced for using ab22492 for IHC-P. However, the application notes for ab22492 state that it is unsuitable for IHC-P. I read the Lee et al paper myself and while it states that an Abcam RECA1 antibody was used for IHC-P, they didn't state which one was used. How does Abcam know whether Lee et al used ab22492 in the paper, and not another Abcam RECA1 antibody like ab9774? Thanks! ANSWER: Thank you for contacting us and sorry for the delay of our reply. After looking at our archives, we are confident that Lee et al. ordered andused ab22492, not ab9774. Regarding the IHC, the publication mention a use of the antibody in IHC-P or IHC-Fr, it could be either. Therefore, the datasheet has been updated and the application has been changed on the reference so it just states IHC. I hope this information is helpful to you. Please do not hesitate to contact us if you need any more advice or information. Kind regards, Lauretta Low

Answer

Thank you for your query.

The epitope recognised by ab22492 is reported to be sensitive to formaldehyde fixation (which is commonly used in IHC-P). There are publications which have used this clone successfully in IHC-P but we have never been able to replicate their success in our labs. Because of this, we are currently unable to support the use of this product with paraffin sections.

I can suggest to use ab124258. This is the same antibody, but from a subclone of the producingmyeloma and this antibody is tested and guaranteed for IHC-P.

https://www.abcam.com/index.html?datasheet=124258 (or use the following: https://www.abcam.com/index.html?datasheet=124258).

I hope this information is helpful and wish you good luck with your reseach.

Thank you for contacting us.

ab9774 is a well characterized antibody is fully guaranteed to detect the endothelial cells. Please use the product without any hesitation.

ab22492 is also reactive against vascular endothelium so this product is also an option to try.

Answer

Thank you for your reply and this precision.

I have just realised that ab9774 is not an anti-RECA1 antibody. RECA1 is the clone number of this anti-Endothelial Cell antibody.

The only 2 anti-RECA1 antibodies we have in our catalog are https://www.abcam.com/ab22942 and https://www.abcam.com/ab124258.

I am very sorry for the confusion. I would be pleased to offer you a replacement of ab9774 withone unit of ab22942 or ab124258. If you decide to try ab22942 in IHC-P we would be pleased if you could share the results with us so we could confirm if this antibody is suitable or not in IHC-P.

I look forward to hearing from you with details of how you would like to proceed.
