    Negatively regulates matrix metalloproteinase-9 (MMP-9) by suppressing MMP-9 secretion and by direct inhibition of its enzymatic activity. RECK down-regulation by oncogenic signals may facilitate tumor invasion and metastasis. Appears to also regulate MMP-2 and MT1-MMP, which are involved in cancer progression.

  • HepG2 (human liver hepatocellular carcinoma cell line) cells stained for RECK (green) using ab238162 at 1/100 dilution in ICC/IF, followed by Alexa Fluor 488-conjugated Goat Anti-Rabbit IgG (H+L) secondary antibody.

    The cells were fixed in 4% formaldehyde, permeabilized using 0.2% Triton X-100 and blocked in 10% normal Goat Serum. The cells were then incubated with the antibody overnight at 4°C. Counterstained with DAPI.

  • Paraffin-embedded human kidney tissue stained for RECK using ab238162 at 1/300 dilution in immunohistochemical analysis.

    After dewaxing and hydration, antigen retrieval was mediated by high pressure in a citrate buffer (pH 6.0). Section was blocked with 10% normal goat serum for 30 minutes at RT. Then primary antibody (1% BSA) was incubated at 4°C overnight. The primary is detected by a biotinylated secondary antibody and visualized using an HRP conjugated SP system.
