    • Function

      Essential for bone resorption and osteoclast differentiation (By similarity). Reversible hydration of carbon dioxide. Can hydrates cyanamide to urea. Involved in the regulation of fluid secretion into the anterior chamber of the eye.
    • Involvement in disease

      Defects in CA2 are the cause of osteopetrosis autosomal recessive type 3 (OPTB3) [MIM:259730]; also known as osteopetrosis with renal tubular acidosis, carbonic anhydrase II deficiency syndrome, Guibaud-Vainsel syndrome or marble brain disease. Osteopetrosis is a rare genetic disease characterized by abnormally dense bone, due to defective resorption of immature bone. The disorder occurs in two forms: a severe autosomal recessive form occurring in utero, infancy, or childhood, and a benign autosomal dominant form occurring in adolescence or adulthood. Autosomal recessive osteopetrosis is usually associated with normal or elevated amount of non-functional osteoclasts. OPTB3 is associated with renal tubular acidosis, cerebral calcification (marble brain disease) and in some cases with mental retardation.

    I am not sure I understand: the protein may not have a functional
    His-tag nor does it have Carbonic Anhydrase activity and this is fine?
    I am sorry but the info about lack of activity is not listed under
    product description- i.e. you are selling a protein that you know is
    inactive and you are not sharing this info in product
    description? At least there should be a note additional info under
    "Scientific Support". Sorry but this is not obvious (nor expected)
    that one can sell inactive protein and assume this is fine.

    Upon re-reading your first email I did realize that you state the protein did not bind the NTA chip, which if it is His tagged it certainly should bind this chip. I apologize for this oversight. For this reason I am happy to offer a free replacement, credit or refund on your purchase of ab87351.You do not need to return the unused portion.

    Please let me clarify as well that I did not mean to suggest we know this protein is not active. We just have no functional data to suggest whether this protein is or is not active. Functionality has not, to our knowledge,been tested. We would describe the protein as 'active' if it had tested positive in a functional assay.

    product: ab87351 Carbonic anhydrase His-tagged

    I purchased this protein to serve as a positive control for SPR experiment on an NTA-chip. This protein had not been captured on the NTA surface and whatever little was captured was inactive.

    I have a positive control- CAII bovine from sigma, I know it binds the small molecules that I work with, while the His-tagged protein from your source remained completely inactive.

    I would like to return this product as this protein is dead in CA activities. please provide me with RMA and shipping instructions.


    PS. your shipping label is printed with such a tiny print (I am guessing it is size 65 or smaller) that is virtually unreadable without magnifying glass.

    Thank you for your enquiry.

    Thus far the only data we have regardingab87351is that the protein is the correct amino acid sequence and a protein of the expected molecular weight can be purified and stained via commassie. This Carbonic Anhydrase II recombinant protein has not previouslybeen tested for activity or any other functional properties and as such we cannot guarantee functional activity from this product.

    Also please note that a similar question regarding potential of ab87351 to have activity, asked in March 2012,is recorded under the 'Scientific Support' tab onour online product datasheet. Please see the text of the question and answer below:

    1) Do you have a protocol and buffer information for using this His-tagged protein on a Ni++ column?

    - We regrettably do not share our purification protocol, but the buffer used is 20 mM Tris pH8.0, 0.5M NaCl, 5mM imidazole.

    2)Assumablya Ni++ column was used to purify the protein. What conditions were specifically used?

    - No special conditions were used during the purification.
