    Question

I do not understand the meaning of "specific activity is < 1 units/ml".
When we use it, we need assess the activity again. As we lack of
experience of ALDH activity assay, could you let us know the protocol you
used for the assessment? Thanks again for the help in advance.

    Answer

Thank you for your reply. The specific information activity for these products is as follows:

    ab87415: Specific activity is > 0.14 units/ml

    ab93463 and ab117154: Specific activity is > 0.5 units/ml

The protocol used to measure the specific activity is:

    Activity Assay

    1) Prepare a 3ml reaction mixture into a suitable container: The final concentrations are 103 mM Tris, 0.67 mM beta-NAD, 100 mM potassium chloride, 10 mM 2-mercaptoethanol, 2 mM acetaldehyde, 0.0007 % (w/v) BSA.
    2) Equilibrate to 25C and monitor the A340nm until value is constant using a spectrophotometer.
    3) Add 5ug of recombinant ALDH2 into reaction mixture and mix immediately.
    4) Record the increase in A340nm for 5 minutes

I hope this helps, please let me know if you need any additional information.
