    AMPK (5'-prime-AMP-activated protein) is a heterotrimer of an alpha catalytic subunit, a beta and a gamma non-catalytic regulatory subunits. It belongs to the ser/thr protein kinase family. AMPK is a cellular energy sensor conserved in all eukaryotic cells. The kinase activity of AMPK is activated by the stimuli that increase the cellular AMP/ATP ratio. AMPK regulates the activities of a number of key metabolic enzymes through phosphorylation. It protects cells from stresses that cause ATP depletion by switching off ATP-consuming biosynthetic pathways. AMPK is also responsible for the regulation of fatty acid synthesis by phosphorylation of acetyl-CoA carboxylase. It regulates cholesterol synthesis via phosphorylation and inactivation of hydroxymethylglutaryl-CoA reductase and hormone-sensitive lipase.

Please find attached the protocol for the AMPK activity assay. You can substitute your substrates for the one mentioned in the protocol using the same concentration, 1mg/ml

You will note that there is no concentration specified in the protocol for the working solution of AMPK, only a volume of 10ul. The amount of AMPK required is lot-dependent. Our current lot, GR11611, has an activity of 737 nmol/min/mg. We recommend testing a range of protein amounts, but please note that the current datasheet graph is for a different lot that had over twice the activity of the current lot. If you decide to choose only one amount, I suggest using a minimum of 5ng/10ul for lot GR11611.

I have been in contact with the lab regarding concentration data for ab79803 as you have requested. Their response is below:

Our AMPK(A1B1G1) had a total of 0.1ug/ul concentration, so A1 (kinase domain) had ˜0.033ug/ul in the prep and it’s calculated MW is ˜63630Da, so it has ˜0.52uM concentration.
