    Controls the classical pathway of complement activation. It binds as a cofactor to C3b/C4b inactivator (C3bINA), which then hydrolyzes the complement fragment C4b. It also accelerates the degradation of the C4bC2a complex (C3 convertase) by dissociating the complement fragment C2a. It also interacts with anticoagulant protein S and with serum amyloid P component. The beta chain binds protein S.

Application
Functional Studies
We analyzed the functionality of Human C4 binding protein full lenght protein,: the protein's ability to act as a cofactor for the C3b/C4b inactivator which then hydrolyzes the complement fragment C4b.
Therefore we incubated hirudinized (to preserve an intact complement system) human whole blood samples with 'double' physiological concentrations by the addition of a second physiological amout of C4-BP (200µg/ml). Samples were further mixed with 'Compound X' and incubated for 30min at 37°C, rotating. Consecutively we analyzed an antibody-mediated binding ability of 'Compound X' to human erythrocytes, which is assumed to depend (beyond other mechanisms) on opsonization by C4b and subsequent binding to complement receptor 1. This was done by centrifugation of the cell fraction and separate analysis of the 'Compound X' DNA content in the supernatant and the cell pellet (DNA isolation and RealTime PCR DNA contents were normailized using actin). We found a reduced binding of about 30%. Thus, we conclude that the recombinant protein C4-BP is functionally active.
