Answer

Thank you for your reply. I will be helping you while Carolyn is out of the office.

I would imagine that you could run non-reducing conditions, though we haven't specifically tested this. If you're going to be detecing the protein with an antibody, it will depend on the binding of the antibody.

We haven't heard any other complaints of multiple bands while using this protein, but care should be taken to minimize any degradation possibilities.

I hope this information helps. Please contact us with any other questions.

Application
Western blot

Review text: We used this HA-tagged CARHSP1 protein only as a control to quantify our own HA-tagged proteins on a Western blot. For this it worked well, although there are weak degradation products as well as the main 44 kDa band for CARHSP1.
Sample: Human Recombinant protein
Loading amount: 0.3 µg
Treatment: diluted in 0.8M Tris, 0.2M sorbitol, 1% betamercaptoethanol, with bromophenol blue. Added 1% SDS then boiled for 5 min.
Gel Running Conditions: Reduced Denaturing (15% acrylamide gel)
Blocking step: Milk as blocking agent for 18 hour(s) and 0 minute(s) · Concentration: 5% · Temperature: 10°C

Other product details
Dilution: 1/100
Incubation time: 1 hour(s) and 0 minute(s) · Temperature: 20°C · Diluent: TBS + 0.1% tween

Primary antibody (in addition to 'CARHSP1 protein')
Name: Non-Abcam antibody was used: anti-HA
Host species: Rabbit
Dilution: 1/2000

Secondary antibody
Name: Non-Abcam antibody was used: anti-rabbit ECL
Clonality: Polyclonal

Answer

Thank you for your email and your patience.

The producers of this protein confirmed that these bands are degradation products. Unfortunately we do not have another batch without these bands.I am sorry for this inconvenience.

Please see here the comments from the originator on the image in the attachment andseen on ourdatasheet: "The three bands present on the gel are as follows – 44 kDa GST-HA-CRSPH24 (full length), 35 kDa and 30 kDa are both degraded forms for the GST-HA-CRSPH24. I have attached a copy of the SDS-Page gel for this protein and you can clearly see the three bands and it corresponds with the anti-HA western blot.We haven't calculated the appropriate proportion of the full length to the breakdown, but the full length is clearly the majority.Unfortunately we do not have a batch of the protein without these degradation fragments."

I hope you will be able to use this protein nevertheless to quantify your results. In order to apologize for the inconvenience, I can offer a 50% discount on the purchase ofa second unit of this protein. Please let me know if this would be an interesting option for you.

Thank you also for submitting an Abreview. We look forward to receive your feedback!
