Stability and Storage
Shipped at 4°C. Store at +4°C short term (1-2 weeks). Add 1% BSA for extra stability. Upon delivery aliquot. Store at -20°C. Avoid freeze / thaw cycle.
pH: 3.00
Constituents: 0.15% Sodium citrate, 0.58% Sodium chloride, 0.01% DTT -
ReconstitutionCentrifuge the vial prior to opening. Reconstitute in water to a concentration of 0.1-1.0 mg/m. Do not vortex. For extended storage, it is recommended to further dilute in a buffer containing a carrier protein (example 0.1% BSA) and store in working aliquots at -20°C to -80°C. Lyophilized protein is stable at room temperature for 1 month and at 4°C for 6 months. Reconstituted working aliquots are stable for 1 week at 2°C to 8°C and for 3 months at -20°C to -80°C.

Relevance
Acts as a negative regulator of the proliferation of normal cells by interacting strongly with CDK4 and CDK6. This inhibits their ability to interact with cyclins D and to phosphorylate the retinoblastoma protein. Heterodimer with CDK4 or CDK6. Predominant p16 complexes contained CDK6. Interacts (isoforms 1,2 and 4) with CDK4 (both 'T-172'-phosphorylated and non-phosphorylated forms); the interaction inhibits cyclin D-CDK4 kinase activity. Interacts with ISCO2. Widely expressed but not detected in brain or skeletal muscle. Isoform 3 is pancreas-specific. -
