Thank you for your call earlier this week and for your patience while I have been in touch with the lab that produces this protein. My contact at the production lab did not have any specific recommendations, and they unfortunately do not have any data for the protein in ELISA so I have removed this as a tested application.I would still expect this protein to be detected by antibodies diluted in either of the buffers that you mentioned. I would recommend trying a couple of assays to determine the source of the problem: 1) Run a quick dot blot to make sureeach individual antibody reacts with the protein. We have a protocol for this kind of assay- If the antibodies individually react with the protein, but there is no signal in the sandwich, the binding sites of the antibodies might interfere with each other in some way. Since the conformation of this recombinant protein could be different than the native conformation of the protein in your samples, this won't necessarily be seen with your samples. If you do find that this is the case, you can try a different pair of antibodies, though I realize you've already developed the sandwich pair with these particular antibodies. If you can find a protein that has been isolated from native tissue, it may also work better in this particular assay. 2) The protein should be at the stated concentration, but itwould be a good idea to check the concentration of the working dilutions by Nanodrop or BCA assay, etc. As we discussed, I would be happy to send another vial of the protein so you can continue to troubleshoot.
