  • Function

    Has both glyceraldehyde-3-phosphate dehydrogenase and nitrosylase activities, thereby playing a role in glycolysis and nuclear functions, respectively. Participates in nuclear events including transcription, RNA transport, DNA replication and apoptosis. Nuclear functions are probably due to the nitrosylase activity that mediates cysteine S-nitrosylation of nuclear target proteins such as SIRT1, HDAC2 and PRKDC (By similarity). Glyceraldehyde-3-phosphate dehydrogenase is a key enzyme in glycolysis that catalyzes the first step of the pathway by converting D-glyceraldehyde 3-phosphate (G3P) into 3-phospho-D-glyceroyl phosphate.

The datasheet states the protein amount is 500ug and should be diluted in 260ul of either HEPES,TRIS or distilled water. Could you please specify regarding desired bufferpH and concentrations and what are the considerations for choosing a buffer?

Answer



We would recommend reconstituting the GAPDH protein (Active), ab82633, with 260 uL of distilled sterilised water. This will produce a buffer of 20mM HEPES, pH 7.0. 10% glycerol can be added if longer term storage is required.
260µl is the volume used when the protein is aliquoted before being lyophilised. Using 260µl will bring the protein concentrationto 2 mg/ml.

The GAPDH protein ab82633 and ab77109 are both the full length GAPDH expressed in E. coli, however only ab82633 has been tested for its enzymatic activity. The specific activity was found to be 35.7 U/mg. One unit of activity of GAPDH is defined as the amount of enzyme required to convert 1 µmole of NAD to NADH per minute under standard assay conditions.
